狐狸源肺炎克雷伯氏菌分离株生物学特征分析 被引量:10

摘要 为确定狐狸源肺炎克雷伯氏菌的致病性、耐药性、耐药机制及其生物被膜(BF)形成能力,本研究对分离自病死狐狸的10株肺炎克雷伯氏菌进行形态特征、培养特性、生化试验、16S r RNA鉴定及khe基因鉴定,利用K-B法检测其药敏性,利用PCR方法检测了其耐药基因、荚膜血清型、毒力基因,利用96孔微量板测定BF形成能力并进行了分离菌对小鼠的致病性试验。结果显示,这10株肺炎克雷伯氏菌均对多种常用抗菌药物耐药,均可以引起小鼠发病或死亡,但BF形成能力有明显差异。部分菌株携带floR、mcr-1、TEM等耐药基因和wabG、fimH、uge、rmpA等毒力基因,89F-2、58F-2、60N-1属于K1血清型。本研究结果为狐狸源肺炎克雷伯氏菌病的检测、诊断及防治提供了实验依据。同时,本实验首次从狐狸源肺炎克雷伯氏菌中检测到粘菌素耐药基因mcr-1,这将为黏菌素的临床使用和风险评估提供技术参考。 To analyze the characteristics of the pathogenicity,drug resistance,resistance mechanism and the ability of biofilm formation of the Klebsiella pneumonia isolated from foxes,ten K.pneumonia were isolated from die foxes and identified by morphology,culture characteristics,biochemical test,16S rRNA and khe gene amplification.The drug resistance,the resistant genes,biofilm formation ability and the pathogenicity in mice of the isolates were further examined using the K-B method and PCR,etc.The results showed that all the isolates of K.pneumonia were resistant to a variety of commonly used antimicrobial agents,all of which could lead to the onset or death of mice,but there was a significant difference in biofilm formation ability. Some isolates carried floR,tact-1,TEM and virulence genes,such as wabG,fimH,uge,rmpA.The isolates of 89F-2,58F-2and 60N-1 belonged to K1 serotype.The results provided experimental basis for the detection,diagnosis,treatment and prevention of K.pneumonia.At the same time,this study first detected the mcr-1gene from K.pneumonia,which would provide a technical reference for the clinical use and risk assessment of mycosin.
