山腊梅叶中8种成分含量测定及主成分分析 被引量:13

摘要 目的建立同时测定山腊梅叶中8个成分(东莨菪苷、东莨菪内酯、异嗪皮啶、山柰酚-3-O-芸香糖苷、紫云英苷、滨蒿内酯、槲皮素、山柰酚)含量的高效液相色谱方法。方法采用Welch-C_(18)色谱柱(250mm×4.6 mm,5μm),以乙腈(A)-0.1%甲酸酸水(B)为流动相,梯度洗脱,流速:0.7 mL·min^(-1),检测波长:359 nm。结果山腊梅叶中8个成分在考察的浓度范围内,浓度与峰面积之间呈良好的线性关系(r>0. 999),回收率在97.75%~100.12%范围内,RSD均小于3.0%。主成分分析可直观地把江西和浙江产地的样品进行分类。结论该方法简便、准确,精密度高,重复性好,可用于比较不同来源山腊梅叶的差异性及其质量控制。 Objective To establish a RP-HPLC method for simultaneous determination of 8 components(scopolin,scopoletin, isofraxidin, kaempferol-3-O-rutinoside, astragalin, scoparone, quercetin, and kaempferol) in Chimonanthus nitens leaf(CNOL). Methods The separation was performed on a Welch column(250 mm × 4.6 mm,5 μm) with gradient elution using acetonitrile(A) and 0.1% formic acid(B) as mobile phase at a flow rateof 0.7 mL·min-1. The detection wavelength was 359 nm. Results Linearities of all calibration curves were good(r>0. 999) over the tested ranges. The average recoveries were ranged from 97.75% to 100.12% with RSD value below3.0%. Principal component analysis can intuitively classify samples from Jiangxi and Zhejiang origins. Conclusion The established method is simple,fast,and with high precision,sensitivity and repeatability,which can be usedfor comparing the differences of different sources,and the quality control of Chimonanthus nitens Oliv. leaf.
