Optimization of ethanol extraction for Knee and Arthralgia Herbal Paste by central composite design-response surface method

目的优选膝痹膏最佳提取的工艺。方法以阿魏酸、藁本内酯、秋水仙碱的含量及浸膏得率为评价指标,以溶剂倍数、提取时间、乙醇体积分数为主要影响因素,采用星点设计-效应面法优选膝痹膏提取工艺的最佳条件。结果确定膝痹膏的最佳提取工艺为加11倍55%的乙醇回流提取两次,每次2. 5h。结论优选的膝痹膏提取工艺,方法简便、重现性、可预测性较好。

Objective To optimize extraction process of Knee and Arthralgia herbal paste. Methods The extraction effect was evaluated with the content of ferulic acid,ligustilide,colchicine and the extract yield. The main influential factors of extraction techniques included the liquid-solid ratio,extraction time and ethanol volume fraction. The optimal extraction techniques of Knee and Arthralgia herbal paste was selected by central composite design—response surface method. Results The best extraction conditions were as follows: 11-fold of 55 percent ethanol,extracted for twice,2. 5 hour for each time. Conclusion The extraction techniques of Knee and Arthralgia herbal paste is simple,with good reproducibility and predictability.
