摘要
目的建立滇黄精组织培养体系快繁技术体系。方法以云南省文山市的滇黄精种子为试验材料,采用常规组织培养技术,进行种子萌发、不定芽增殖、生根培养基的筛选。结果滇黄精种子萌发采用不附加任何激素的1/2MS培养基较为适宜;根状茎在MS+BA 4. 0 mg/L+NAA 0. 2 mg/L+蔗糖30 g/L+琼脂6. 5 g/L培养基上不定芽的增殖系数最大,达到3. 85;再生植株生根较容易,在MS+NAA 0. 5 mg/L+蔗糖30 g/L+琼脂6. 5 g/L培养基上能达到很好的生根效果。结论研究结果为滇黄精规模化、工厂化育苗、资源保存和育种工作提供理论和技术支撑。
Objective To establish the system of tissue culture and rapid propagation of Polygonatum kingianum.Methods The seeds of P.kingianum from Wenshan ,Yunnan Province were used as experimental materials.The media of seed germination,ad- ventitious bud proliferation and rooting were screened by conventional tissue culture technique.Results 1/2MS medium without any hormone was suitable for seed germination.The adventitious buds multiplication ratio 3.85was achieved on the medium of MS +BA 4.0mg/L +NAA 0.2mg/L +sucrose 30g/L +agar 6.5g/L.Tissue cultured plantlets root easily and the medium composition of MS +NAA 0.5mg·L-1 +sucrose 30g/L +agar 6.5g/L was good for rooting.Conclusion The results can provide the theoretical and technical support for mass seedling production,germplasm resources preservation and breeding of P.kingianum.
