摘要 目的:通过超高效液相色谱-四级杆串联飞行时间质谱(UPLC-Q-TOF),对金钗石斛的乙酸乙酯萃取部位的主要化学成分进行定性分析研究。方法:采用UPLC C_(18)(100 mm×2.1 mm,1.6μm)色谱柱,以乙腈-0.1%甲酸溶液为流动相梯度洗脱;流速0.4 mL/min;柱温30℃。质谱分析采用电喷雾(ESI)离子源,独立数据采集(IDA)、正负离子模式下分别采集数据。针对保留时间、测得的精确相对分子质量,应用MassHunter软件Formula Calculator等功能、天然产物数据库,分析比较各色谱峰的一级二级质谱裂解碎片。结果:从金钗石斛的乙酸乙酯萃取部位共鉴定了45个天然化合物,其主要化学成分包括生物碱类、联苄类、菲类、倍半萜类等。结论:该方法准确、可靠,适用于金钗石斛化学成分的鉴定研究,为金钗石斛的药效物质基础、资源开发利用提供参考。 Objective:To analyze the main chemical constituents of ethyl acetate extract of Dendrobium nobile by UPLC-Q-TOF qualitatively.Methods:UPLC C18(100 mm×2.1 mm,1.6 μm)column was applied with gradient elution of acetonitrile-0.1% formic acid solution,at a flow rate of 0.4 mL/min,and column temperature was 30 ℃.MS analysis was based on ESI ion source,independent data acquisition(IDA),positive and negative ions were respectively collected.Through retention times,measured accurate relative molecular mass,one-level and two-level mass fragments of every chromatographic peak were compared by Formula Calculator function and so on from MassHunter software,natural product databases.Results:There were 45 natural compounds identified from ethyl acetate extract of Dendrobium nobile,the main chemical constituents included alkaloids,bibenzils,phenanthrenes and sesquiterpenes.Conclusion:The method is accurate,reliable,suitable for identification of chemical constituents in Dendrobium nobile,it provides a reference for material basis,development and utilization for resource of Dendrobium nobile.
