黑龙骨对CIA大鼠炎症反应的调节作用机制 被引量:6

摘要 目的:研究黑龙骨水溶性成分对CIA大鼠炎症反应的调节作用及机制。方法:随机选取雄性SD大鼠分为正常对照组、模型组、尼美舒利阳性对照组及黑龙骨高(20 g/kg)、低(10 g/kg)剂量组。除正常对照组外,其余各组复制CIA模型。给药6 w后免疫组化法和免疫蛋白印迹法检测脾和滑膜TAK-1、TAB-1、NF-κBp65蛋白表达,ELISA检测血清MCP-1、CXCL-1、MIP-1含量。结果:与模型组比较,免疫组化显示黑龙骨高剂量组脾组织TAK-1、TAB-1、NF-κBp65蛋白表达显著降低,免疫蛋白印迹显示黑龙骨高剂量组脾和滑膜内TAK-1、TAB-1、NF-κBp65蛋白表达显著降低;ELISA显示黑龙骨高剂量组血清MCP-1、CXCL-1含量显著降低。结论:黑龙骨水溶性成分可能通过调节TAK-1、TAB-1、NF-κBp65、MCP-1、CXCL-1蛋白对CIA模型大鼠炎症反应起到抑制作用。 Objective:To study the regulation and mechanism on inflammatory reaction of water soluble components of Periploca forrestii on CIA rat model.Methods:Male SD rats were randomly selected and divided into normal control group,model group,nimesulide group and high-dose(20 g/kg) and low-dose(10 g/kg) group of Periploca forrestii.CIA model was constructed in the other groups except the normal control group.After six weeks of continuous administration,immunohistochemistry and western-blot were used to detect the expression of TAK-1,TAB-1,NF-κBp65 protein in spleen and synovial membrane.ELISA method was used to detect the content of MCP-1,CXCL-1,MIP-1.Results:Compared with the model group,immunohistochemistry showed that the TAK-1,TAB-1,NF-κBp65 protein expressions were decreased in high-dose group of Periploca forrestii.Western-blot showed that the TAK-1,TAB-1,NF-κBp65 protein expression in spleen and synovial membrane were decreased in high-dose group of Periploca forrestii.ELISA showed that the contents of MCP-1 and CXCL-1 were down significantly in high-dose group of Periploca forrestii.Conclusion:The water soluble components of Periploca forrestii may inhibit the inflammatory response on CIA rat model by regulating the expressions of TAK-1,TAB-1,NF-κBp65,MCP-1 and CXCL-1 protein.
