藏药材白花刺参总皂苷的抗炎作用及机制 被引量:3

Anti-inflammatory effects and mechanism of total saponins of Tibetan medicine Morina nepalensis var. alba

摘要 目的:探讨藏药材白花刺参总皂苷的体内外抗炎作用及机制。方法:采用二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足肿胀以及小鼠棉球肉芽肿炎症模型评价白花刺参总皂苷体内抗炎作用。采用LPS诱导RAW264.7巨噬细胞炎症反应模型,评价白花刺参总皂苷体外抗炎作用。结果:与模型组相比,白花刺参总皂苷40和80mg/kg能够显著抑制二甲苯所致小鼠耳肿胀、角叉菜胶致大鼠足跖肿胀及小鼠棉球肉芽肿;在体外实验中,白花刺参总皂苷20和40μg/ml能够明显抑制炎症介质NO生成,此抑制作用可能是通过下调炎症蛋白iN OS和COX-2蛋白表达水平发挥作用。结论:白花刺参总皂苷在体内外显示了明显的抗炎活性,该结果首次为白花刺参在藏医药中的合理应用提供实验药理学基础,为进一步开发利用白花刺参药材提供科学依据。 Objective:To investigate the anti-inflammatory effects of total saponins of M^nepalensis vat.alba in vivo and vitro.Methods:The anti-inflammatory effects of total saponins of M.nepalensis vat.alba on xylene induced ear edema in mice,carrageenan-induced rat paw edema and granuloma inflammation model in mice were investigated.LPS-induced RAW264.7macrophages eel1inflammation model with different concentrations was also used to evaluate the anti-inflammatory effects of total saponins in vitro.Results:Compared with the control group,total saponins treatments (40and 80mg/kg)significantly inhibited xylene-induced ear edema in mice,carrageenan gum-induced rat paw edema,granuloma in mice.Total saponins at 20and 40μg/ml can inhibit NO production in LPS-indu'ced RAW264.7and inhibit iNOS and COX-2protein expression in vitro.Conclusion:The total saponins showed significant anti-inflammatory activity in vivo and vitro.The results offers pharmacological basis of rational use in Tibetan medicine and provide a scientific basis for the further development and utilization of M. nepalensis var.alba.
