半枝莲总黄酮对高脂高糖糖尿病大鼠模型糖脂代谢及血液流变学的影响 被引量:9

Effect of total flavonoids from Scutellaria barbata on glycolipid metabolism and hemorheology in high glucose and high fat diet induced biabetic rats

摘要 目的:观察半枝莲总黄酮对高脂高糖糖尿病大鼠糖脂代谢及血液流变学的影响。方法:采用喂养高糖高脂饲料加舌下静脉注射25mg/kg链脲佐菌素造糖尿病大鼠模型,大鼠随机分为空白组、模型组、二甲双胍组(250mg/kg)、半枝莲总黄酮400mg/kg、200mg/kg、100mg/kg组,连续给药30天。于给药的第10、20、30天测空腹6小时血糖值;第31天,禁食后12h,灌胃后2h,腹主动脉采血,检测血液流变学的各项指标,并测血清中低密度脂蛋白、高密度脂蛋白、胆固醇、甘油三酯、胰岛素、胰岛素抗体、糖化血清蛋白等指标;取胰腺组织并采用HE染色观察病理变化。结果:与模型组比,二甲双胍组、半枝莲总黄酮各剂量组均可显著降低血糖水平;降低低密度脂蛋白、胆固醇、甘油三酯含量,可显著升高高密度脂蛋白;显著升高胰岛素含量,降低胰岛素抗体和糖化血清蛋白水平,降低全血低切、中切、高切粘度、血浆粘度、红细胞压积、血沉水平;并能显著改善糖尿病模型大鼠的胰岛细胞萎缩。结论:半枝莲总黄酮对大鼠糖尿病模型具有保护作用,可改善大鼠糖尿病模型的糖脂代谢和血液流变学。 Objective:To observe the effects of total flavonoids of Scutellaria barbata on high fat and high glucose in diabetic rats model of glycolipid metabolism and hemorheology.Methods:A diabetic rat model was established by high glucose and high-fat diet with sublingual intravenous injection of 25 mg/kg streptozotocin.The rats were randomly divided into control group,model group,melbine group(250 mg/kg),total flavonoids of Scutellaria barbata groups on 400 mg/kg,200 mg/kg and 100 mg/kg doses,administered for 30 days.The 6 hours fasting blood glucose level was measured on the 10 th,20 th and 30 th day separately after administration.On 31 th days,blood was collected from the abdominal aorta 2 hours after administration,to detect the indexes of hemorheology,the serum low density lipoprotein,high density lipoprotein,cholesterol,triglycerides,insulin,insulin antibody and glycosylated serum protein index.Pathological observation was taken on pancreatic tissue by HE staining.Results:Compared with model group,the blood glucose level,content of low density lipoprotein,cholesterol and triglyceride was significantly reduced in melbine group,total flavonoids of Scutellaria barbata groups on different doses The content of insulin was significantly increased while the levels of insulin antibody and glycosylated serum protein were reduced.Also the value of blood viscosity,plasma viscosity,hematocrit and erythrocyte sedimentation rate was reduced.The atrophy of islet cells in diabetic rats was significantly improve.Conclusion:The total flavonoids of Scutellaria barbata has protective effect on diabetic rats,and it can improve the glycolipid metabolism and hemorheology in diabetes rat model.
