基于LBP/CD14/FOS信号通路探讨芪藤消浊颗粒治疗慢性肾小球肾炎的作用机制 被引量:3

Effect of Qiteng Xiao Zhuo Granule on chronic glomerulonephritis(CGN) rats based on LBP/CD14/FOS signal pathway:a mechanism exploration

摘要 目的:观察芪藤消浊颗粒对慢性肾小球肾炎(CGN)大鼠的LBP/CD14/FOS信号通路的作用。方法:将雄性SD大鼠随机分为正常组、模型组、芪藤消浊颗粒5.4g/kg、10.8g/kg、21.6g/kg组、阳性药黄葵胶囊1.0g/kg组,每组8只。除正常组外,其余各组第1天按3.5 mg/kg,第14天按3.0 mg/kg尾静脉注射阿霉素,复制慢性肾小球肾炎(CGN)大鼠模型。造模后第7天灌胃给予芪藤消浊颗粒及黄葵胶囊,正常组和模型组给予等量溶媒,连续30天。采用HE染色、RT-PCR技术、Western blot检测肾组织中LBP、CD14、p-JNK、p-P38 mRNA和蛋白的表达。结果:芪藤消浊颗粒10.8、21.6g/kg组能显著降低大鼠24h尿蛋白、血清尿素氮(BUN)和肌酐(Cr)的含量;各剂量显著降低CGN大鼠肾脏组织中LBP、CD14、JNK、P38 mRNA的表达,并显著降低CGN大鼠肾脏组织中LBP、CD14、p-JNK、p-P38蛋白表达。结论:芪藤消浊颗粒对CGN大鼠有一定的治疗作用,其机制可能与调节肾脏组织中LBP/CD14/FOS信号通路有关。 Objective:To study the effect of Qiteng Xiao Zhuo Granule on LBP/CD14/FOS signaling pathway in chronic glomerulonephritis(CGN)rats model.Methods:SD rats were randomly divided into six groups(n=8),including normal group,model group,Qiteng Xiao Zhuo Granule(21.6 g/kg,10.8 g/kg,5.4 g/kg),and Huangkui capsule(1.0 g/kg) group.Adriamycin was injected intravenously via tail in model group,Qi Teng Xiao Zhuo granule and Huangkui capsule group to establish CGN model;3.5 mg/kg adriamycin was to be given on the first day and 3.0 mg/kg on the fourteenth day.After model establishment,rats in the Qi Teng Xiao Zhuo granule and Huangkui capsule group were intragastric administered by Qiteng Xiao Zhuo Granule and Huangkui capsule daily,for 30 days.Pathological changes were observed by Hematein Eosin(HE);The real-time fluorescent quantitative PCR,Western blotting were performed to detect the expressions of LBP,CD14,JNK,P38 mRNA and protein.Results:Compared with model group,the Qiteng Xiao Zhuo Granule not only effectively decreased the contents of 24 h urine protein;BUN and Cr(P<0.01 or P<0.05) and expressions of LBP,CD14,JNK,P38 mRNA(P<0.01 or P<0.05),but also inhibited expressions of LBP,CD14,p-JNK,p-P38(P<0.01 or P<0.05) in the kidney.Conclusion:The Qiteng Xiao Zhuo Granule has certain effect on model of CGN rats,which might be related to decrease the expressions of LBP,CD14,JNK and P38 in the kidney and block LBP/CD14/FOS signaling pathway.
