温针灸对慢性束缚应激大鼠焦虑抑郁样行为及BDNF/TrkB通路的影响 被引量:4

Effect of warm-acupuncture on anxiety-depression-like behavior and BDNF/TrkB pathway in chronic restraint stress rats

摘要 目的探讨温针灸对慢性束缚应激大鼠焦虑抑郁样行为及海马脑源性神经营养因子(brain derived neurotrophic factor,BDNF)/酪氨酸蛋白激酶受体B(tyrosine kinase B,TrkB)通路的影响。方法将SD大鼠随机分为四组:正常组、模型组、温针组、氟西汀组,除正常组外,其余各组均进行束缚应激造模,每天束缚6h,持续21d。温针组于造模前选取“百会”、“印堂”穴进行温针灸治疗,每天1次;氟西汀组于造模前灌胃给予抗抑郁药氟西汀,浓度为10mg/kg。造模结束后采用高架十字迷宫和强迫游泳实验分别评价大鼠的焦虑和抑郁样行为,酶联免疫法检测海马单胺递质5-羟色胺(5-hydroxytryptamine,5-HT)、多巴胺(dopamine,DA)含量,免疫组化法检测海马CA1区BDNF、TrkB阳性细胞数,Western-blotting法检测海马BDNF、TrkB蛋白表达情况。结果与正常组比较,模型组大鼠焦虑和抑郁样行为显著,海马单胺递质含量显著下降(均P<0.05),BDNF、TrkB阳性细胞数及蛋白表达均显著下降;在温针灸治疗后,模型大鼠的焦虑和抑郁样行为得到缓解,海马单胺递质含量上升,BDNF、TrkB阳性细胞数及蛋白表达均明显回升(P<0.05)。结论温针灸能显著改善慢性束缚应激大鼠焦虑和抑郁样行为,升高脑内单胺递质含量,其机制可能与调控BDNF/TrkB通路有关。 Objective To investigate the effects of warm-acupuncture on anxiety-depression-like behavior and BDNF/TrkB pathway in hippocampus of rats with chronic restraint stress.Methods SD rats were randomly divided into normal,model,warm-acupuncture and fluoxetine group.In addition to normal group,the rest of the groups were modeled by restraint stress ,bound for 6hours a day for 21days.The warm-acupuncture group selected "Baihui"and "Yintang"points for warm acupuncture moxibustion before modeling,once a day;In fluox- etine group,the antidepressant fluoxetine was administered by intragastric administration at a concentration of 10 mg?kg-1.At the end of the model,the rats were evaluated for anxiety and depression-like behavior using elevated plus maze and forced swimming test.The levels of 5-HT and DA in the hippocampus .were determined by enzyme-linked immunosorbent assay.The number of BDNF and TrkB positive cells in hippocampal CA1 region was detected by immunohistochemical method and protein expression was detected by Western-blotting.Results Compared with normal group,the anxiety and depression-like behaviors were significant in model rats,the content of monoamine neurotransmitters in hippocampus was significantly decreased,the number of BDNF,TrkB positive cells and protein expression were significantly decreased (all P <0.05).After the warm-acupuncture treatment,the anxiety and depression-like behaviors of model rats were alleviated,content of monoamine neurotransmitters in hippocampus increased,and the number of BDNF,TrkB positive cells,and protein expression all increased significantly (all P <0.05).Conclusion Warm -acupuncture could significantly improve anxiety and depression-like behavior in rats with chronic restraint stress and increase the content of monoamine transmitters in the brain,which might be related to the regulation of BDNF/TrkB pathway.
