摘要 目的:探讨口服鹿茸蛋白提取物(pilose antler extracts,PAEs)对急性心梗小鼠模型的保护作用。方法:采用C57BL/6小鼠冠状结扎方法制备急性心梗动物模型,将小鼠按随机数字表法分为假手术组、模型组、氯沙坦钾组、PAEs高、中、低剂量组,造模成功后灌胃给药,14 d后N-BT染色法计算心肌梗死面积,HE染色法观察心肌损伤程度,Masson染色法确定纤维含量并确定心肌酶含量的变化。结果:PAEs低、中、高浓度对小鼠心肌缺血程度(Σ-ST)有显著改善作用,显著减小梗死面积,降低纤维化程度和小鼠心梗后心肌组织CK、CK-MB、LDH与血清LDH含量。结论:口服鹿茸蛋白提取物对损伤心肌具有明显的保护作用,其作用机制可能与调节心肌酶水平、降低心肌纤维化程度、促进缺血心肌恢复等机制有关。 Objection: To observe the cardioprotective effects of proteins from Pilose antler extracts( PAEs) on acute ischemic myocardial injury in mice. Method: The model mice of acute myocardial ischemia injury was produced by ligaturing the coronary attery( CA),and then mice were randomly divided into 6 groups as below: sham group,model group,low/medium/high-dose PAEs group and the positive control group( n = 10). The ischemic size,ST elevation,the levels of CK,CK-MB,LDH in serum and myocardial tissue were measured in mice at 14 days after CA occlusion. Results:Treatment with PAEs at 60 and 180 mg/kg significantly decreased ST elevation compared with the MI mice. Treatment of PAEs at dose 60,120 and 180 mg/kg resulted in reductions in the infarct size and PAEs also significantly decreased levels of CK,CK-MB,LDH in myocardial tissue,and decreased LDH level in serum. Conclusion: PAEs exerts significant cardioprotective effects against acute ischemic myocardial injury in mice,the mechanism may be related to regulating enzyme levels,reducing myocardial fibrosis and promoting ischemic myocardial recovery.
