肾透明细胞癌组织GOLPH3表达临床意义

Expression and clinical significance of GOLPH3 in clear renal cell carcinoma tissues

摘要 目的高尔基蛋白3(Golgi phosphoprotein 3,GOLPH3)与多种肿瘤的发生相关,在肿瘤组织中高表达,本研究拟探讨GOLPH3在肾透明细胞癌(renal clear cell carcinoma,RCCC)组织中的表达及其临床意义。方法采用免疫组化SP法检测72例RCCC组织和40例癌旁正常组织中GOLPH3的表达,采用蛋白质印迹法检测10例RCCC组织和癌旁正常组织中GOLPH3的蛋白表达。结果RCCC组织GOLPH3阳性表达率(53%)高于癌旁正常组织(20%),χ^2=38.798,P<0.05;GOLPH3蛋白高表达患者的平均生存期(52.34个月,95%CI:42.803~61.881)低于低表达者(68.79个月,95%CI:52.984~74.587),χ^2=23.42,P<0.05;多因素Cox回归显示TNM分期(T1~T2/T3~T4)HR=3.76(95%CI:2.87~4.65)、Fuhaman分级(G1~G2/G3~G4)HR=2.63(95%CI:1.98~3.56)、GOLPH3表达水平(HR=2.94,95%CI:2.35~3.58)均是RCCC预后的独立危险因素。结论 GOLPH3在RCCC的发生发展中发挥重要作用,有可能成为新的RCCC检测和监控的指标。 OBJECTIVE Golgi phosphoprotein 3(GOLPH3)is associated with the occurrence of various tumors,and is highly expressed in tumor tissues.This study was to investigate GOLPH3 expression in renal clear cell carcinoma and its clinical significance.METHODS Immunohistochemical was used in 72 cases of renal clear cell carcinoma and 40 cases of tumor-adjecent tissues.Western bloting was used in 10 cases of renal clear cell carcinoma and adjacent tissues.RESULTS GOLPH3 expression(53%)in renal clear cell carcinoma was significantly higher than that in normal tissues(20%),χ^2=38.798,P<0.05.The mean survival time(52.34 months,95%CI:42.803-61.881)of patients with high GOLPH3 protein expression was lower than that of patients with low GOLPH3 protein expression(68.79 months,95%CI:52.984-74.587),χ^2=23.42,P<0.05.Multivariate Cox regression analysis showed that TNM stage(T1-T2/T3-T4)HR=3.76(95%CI:2.87-4.65),Fuhaman grade(G1-G2/G3-G4)HR=2.63(95%CI:1.98-3.56),GOLPH3 expression level(HR=2.94,95% CI:2.35-3.58)are independent risk factors in renal clear cell carcinoma.CONCLUSION GOLPH3 plays an important role in the development of renal clear cell carcinoma and it may be a new biomarker for renal clear cell carcinoma.
