蛇含委陵菜乙醇提取物的抗炎及抑菌活性研究 被引量:6

摘要 目的对蛇含委陵菜(Potentilla kleiniana)乙醇提取物的抗炎及抑菌活性进行初步研究。方法使用70%乙醇对蛇含委陵菜进行浸泡提取,色谱法测定蛇含委陵菜中总黄酮、总酚及鞣质含量;使用高糖诱导分化后的PC12神经细胞损伤模型,评价蛇含委陵菜乙醇提取物的抗炎活性及其对细胞活性的影响;牛津杯法抑菌实验检测蛇含委陵菜乙醇提取物的抑菌活性。结果蛇含委陵菜乙醇提取物中总黄酮含量为28.9 mg·g^(-1)芦丁当量,总酚含量为11.8 mg·g^(-1)没食子酸当量,鞣质含量为10.8 mg·g^(-1)没食子酸当量。高糖(75 mmol·L-1)孵育分化后PC12神经细胞72 h后细胞内炎症因子肿瘤细胞因子(TNF-α)、白介素(IL)-1β及IL-6的mRNA表达明显增加,细胞培养液中一氧化氮(NO)及前列腺素E2(PGE2)的含量增加,细胞活性明显降低;蛇含委陵菜乙醇提取物呈浓度依赖性地抑制高糖诱导的细胞内炎症因子(TNF-α、IL-1β及IL-6)生成,并降低细胞培养液中NO和PGE2的含量,同时上调细胞活性。蛇含委陵菜乙醇提取物对痤疮丙酸杆菌、大肠埃希菌、金黄色葡萄球菌、枯草芽孢杆菌、白色葡萄球菌、白假丝酵母菌、铜绿假单胞菌及变形杆菌均无明显的抑菌作用。结论蛇含委陵菜乙醇提取物能抑制高糖诱导的分化后PC12神经细胞活性下降,其机制可能与其抗炎活性有关。 Objective To determine the anti-inflammation and antibacterial activity of the ethyl alcohol extractive of Potentilla kleiniana.Methods Ethanol(70%) was used to extract Potentilla kleiniana.The content of total flavonoids, total phenols and tannins in the ethyl alcohol extract of Potentilla kleiniana was determined by chromatography.In the high glucose-induced differentiated PC12 cell damage model, the anti-inflammatory activity of ethyl alcohol extract of Potentilla kleiniana and its effect on the cell activity were evaluated.The antibacterial activity of ethyl alcohol extract of Potentilla kleiniana was tested by Oxford cup method.Results The average content of total flavonoid, total phenol and tannin in the ethyl alcohol extract of Potentilla kleiniana was 28.9 mg·g^-1 rutin equivalent, 11.8 mg·g^-1 gallic acid equivalent and 10.8mg·g^-1 gallic acid equivalent, respectively.The mRNA expression of inflamm atory cytokines TNF-α,IL-1 β and IL-6 in the differentiated PC12 cells and the content of nitr ic oxide and prostaglandin E2 in the culture medium of differentiated PC12 cells increased significantly 72 hours after the differentiated PC12 cells incubated with 75 mmol·L^-1 high glucose.The activity of differentiated PC12 cells decreased significantly 72 hours after 75 mmol·L^-1 high glucose treatment.Potentilla kleiniana ethyl alcohol extract inhibited the mRNA production of inflammatory cytokines(TNF-α,IL-1 β and IL-6) and the content of nitric oxide and prostaglandin E2 in the culture medium of differentiated PC12 cells in a concentration-dependent manner.At the same time, Potentilla kleiniana ethyl alcohol extract up-regulated the viability of differentiated PC12 cells in a concentration-dependent manner.The ethyl alcohol extract of Potentilla kleiniana had no obvious antibacterial activity against Propionibacterium acnes, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Staphylococcus aureus, Candida albicans, Pseudomonas aeruginosa and Proteus.Conclusion Ethyl alcohol extract of Potentilla kleiniana has obvious anti-inflammatory effect on high glucose-induced differentiated PC12 cells.The reversal mechanism of high glucose-induced decrease in differentiated PC12 cell activity by the ethyl alcohol extract of Potentilla kleiniana may be related to its anti-inflammatory effect.
