陕西产香青兰质量标准的探讨性研究

摘要 目的建立香青兰的定性和定量方法。方法参照2015年版《中国药典》附录相关方法,对该药材水分、总灰分、酸不溶性灰分、醇溶性浸出物进行测定;采用薄层色谱法,分别以迷迭香酸为对照,对该药材进行定性鉴别;采用HPLC法以迷迭香酸为指标,通过系统的方法学考察,建立了香青兰中迷迭香酸的含量测定方法。色谱柱为Ermsil C_(18)色谱柱(250 mm×4.6 mm,5μm);以甲醇-0.1%磷酸水溶液(43∶57)为流动相,流速为0.6 mL·min^(-1),柱温35℃,检测波长为210 nm。结果水分平均为8.65%,总灰分平均为9.85%,酸不溶性灰分平均为3.29%,醇溶性浸出物平均6.45%;香青兰薄层色谱,斑点清晰,分离效果理想;HPLC法测得迷迭香酸含量在1.42~3.76 mg·g^(-1)。结论所建立的薄层色谱鉴别和高效液相色谱含量测定方法灵敏,准确可靠,重复性好,为香青兰的质量研究提供了科学依据。 Objective To establish a qualitative and quantitative method to determine Dracocephalum moldavica L..Methods According to related methods in the appendix of the 2015 Chinese Pharmacopoeia, the moisture, total ash, acid insoluble ash and alcohol-soluble extracts of Chinese crude drug were determined.Rosemary acid was used as control in the qualitative identification by thin layer chromatography.Rosemary acid was determined by HPLC after systematic methodological investigation.The chromatographic conditions were as follows: Ermsil C 18 column(250 mm×4.6 mm, 5 μm); methanol-0.1% phosphoric acid water solution(43∶57) as the mobile phase, the flow volumn was 6 mL·min^-1,the column temperature was 35℃,and the wavelength was 210 nm.Results The average moisture was 8.65%, the average ash content was 9.85%, the average acid insoluble ash was 3.29%, and the alcohol soluble extract was 6.45%.The thin layer chromatography was clear with ideal separation.The content of rosmarinic acid was at 1.42-3.76 mg·g^-1.Conclusion Both TLC and HPLC prove sensitive, accurate, reliable and reproducible in the qualitative and quantitative analysis of Dracocephalum moldavica L., which provides a scientific basis for the study of Dracocephalum moldavica L..
