鸡血藤有效部位提取物紫外-可见分光光度法测定总黄酮含量

UV-vis Spectrophotometry Determination for Total Flavonoids from Spatholobus suberectus Dunn. Extract

目的:建立以紫外-可见分光光度法测定鸡血藤有效部位提取物总黄酮含量的方法。方法:以表儿茶素为对照品,以5%亚硝酸钠-10%硝酸铝-氢氧化钠试液为显色体系,在498nm波长下以分光光度法测定总黄酮。结果:表儿茶素浓度在0. 0500~0. 3003mg·ml-1范围内线性关系良好,其线性回归方程为Y=3. 7341X-0. 0238,r=0. 9998 (n=6),精密度RSD为0. 11%,稳定性RSD为2. 20%,重复性RSD为1. 80%,平均加样回收率为97. 86%,RSD为2. 15%(n=9),测得鸡血藤有效部位提取物总黄酮含量为77. 99%。结论:总黄酮含量的紫外-可见分光光度法准确度高、重复性好。

Objective:To establish a ultraviolet-visible spectrophotometry determination for total flavonoids from Spatholobus suberectus Dunn.extract.Methods:Epicatechin as reference substance,5% sodium nitrite-10% aluminum nitrate-4.3% sodium hydroxide solution as chromogenic system,based on 498nm absorption wavelength arid the UV-vis spectrophotometry,to determine the test samples.Results :Epicatechin concentration in the range of 0.05005-0.3003mg ·ml-1 was good,and the linear regression equation was Y=3.738X-0.0239(r=0.9998);RSD of the stability was 2.20%;RSD of the precision was 0.11%;RSD of the repeatability was 1.80%;The average sample recovery rate was 97.65%,RSD was 2.15%(n =9); Measured the total flavonoids conten in test samples was 77.99%.Conclusion:Established the determination for total flavonoids from Spatholobus suberectus Dunn.extract was with high accuracy and good repeatability.
