电刺激对小鼠成纤维细胞L 929机械力损伤的保护作用

Protective effects of electrical stimulation on mechanical stress-induced oxidative damage in mouse fibroblast L 929 cells

摘要 目的探讨电刺激(electrical stimulation,ES)对机械力诱导的小鼠成纤维细胞L 929氧化损伤的保护作用。方法将小鼠成纤维细胞L 929分为正常对照组、电刺激组、加力模型组和加力后电刺激组。采用CCK-8法检测细胞活性,间接化学荧光法检测细胞活性氧(reactive oxygen species,ROS)含量变化,JC-1染色法检测细胞内线粒体膜电位(mitochondrial membrane potential,MMP)变化,分光光度法测定Caspase-3酶活性变化。结果加力组细胞活性明显下降,细胞内ROS增加,线粒体膜电位下降,Caspase-3酶活性增强,差异均有统计学意义(P <0.05)。与加力组相比,加力后电刺激组细胞活性增强,线粒体膜电位水平上升,细胞内ROS含量减少,Caspase-3酶活性减弱,差异均有统计学意义(P <0.05)。结论电刺激可通过维持细胞活性、减少细胞凋亡、减少细胞内ROS含量及Caspase-3酶活性,而对机械力诱导的细胞氧化损伤起到一定的保护作用。 Objective To explore the protective effects of electrical stimulation on mechanical stress -induced oxidative damage in mouse fibroblast L 929 cells.Methods The mouse fibroblast L 929 cells were divided into four groups:blank control group, electrical stimulation group,mechanical stress group and electrical stimulation after mechanical stress group.The cell viability and cellular senescent state were analyzed using CCK -8 .Fluorometric assays were performed to detect the formation of reactive oxygen species (ROS)and mitochondrial membrane potential detection (JC-1)staining in the cells.The caspase-3 activity was detected by Caspase-3 Activity Assay Kit.Results The cell viability in mechanical stress group decreased significantly,intracellular ROS increased,mitochondrial membrane potential decreased,and Caspase-3 enzyme activity increased.The differences were statistically significant (P <0.05).Compared with mechanical stress group,the cell viability in electrical stimulation after mechanical stress group increased,the mitochondrial membrane potential increased,the intracellular ROS content decreased,and the Caspase-3 enzyme activity weakened.The differences were statistically significant (P <0.05).Conclusion Electrical stimulation can maintain cell viability,reduce cell apoptosis,reduce intracellular ROS and Caspase-3 activity..Therefore,the electrical stimulation may be a potential method for the prevention and treatment of mechanical stress - induced cell injury.
