摘要
目的:探讨复发性唇疱疹实热“上火”患者Th17、Treg、CD8、CD19细胞含量及Th17、Treg相关细胞因子变化,从免疫学角度探讨复发性唇疱疹实热“上火”发病机制,为该病防治提供理论基础。方法:临床上收集复发性唇疱疹实热“上火”患者28例和非疱疹人46例(正常平和质26例、非复发性疱疹阴虚和非复发性疱疹实热型10例),采用流式细胞检测仪检测外周血Th17、Treg、CD8、CD19细胞含量;采用Elisa法检测血浆中IL-4、TGF-β、IL-2、IL-10、IL-17A和GM-CSF细胞因子含量。结果:与正常平和质人相比,RHL实热患者外周血淋巴细胞中Th17细胞、CD4^+CD25^+Treg细胞、Foxp3^+Treg细胞含量显著升高(P<0.05);IL-2、IL-10和TGF-β含量显著升高(P<0.05)。与非复发性疱疹阴虚人相比,RHL实热患者Th17细胞和记忆B细胞(CD19^+)含量显著升高(P<0.05),非复发性疱疹实热患者血浆TGF-β显著上升(P<0.05)。与非复发性疱疹实热型相比,检测指标无统计学差异。非复发性疱疹实热外周血B细胞(CD19^+)细胞含量显著高于非复发性疱疹阴虚人(P<0.05)。结论:复发性唇疱疹实热“上火”患者可能与免疫功能紊乱密切相关。
Objectives: To study the changes of Th17, Treg, CD8, CD19 and their related cytokines in recurrent herpes labialis(RHL) patients with excessive heat. Methods: Peripheral blood was collected from 28 patients with excessive heat of RHL and 46 non RHL patients(including 26 gentle quality, deficiency of Yin 10 and excessive heat 10 cases). Flow cytometry was used to detect the percentages of Th17, Treg, CD8, CD19. ELISA assay was conducted to further verify the expression levels of cytokines that were screened by protein microarrays. Results: Compared with gentle quality volunteers, percentages of Th17 cells, CD+4CD+25Treg cells and Foxp3+Treg cells were significantly incresed(P<0.05). IL-2, IL-10 and TGF-β also incresed(P<0.05). Compared with deficiency of Yin volunteers, percentages of Th17 and B cells were significantly incresed(P<0.05)and TGF-β also incresed(P<0.05). Compared with excessive heat volunteers, the levels of B cells in deficiency of Yin volunteers significantly fell behind. Conclusions: Our results indicated that the immunity function was broken in RHL patients with excessive heat.
