棕榈子高效液相色谱指纹图谱的构建

Establishment of HPLC Fingerprint for Fructus Trachycarpi

目的:构建棕榈子的高效液相色谱指纹图谱并应用于不同产地棕榈子的相似性分析。方法:采用80%甲醇水浴回流提取法提取棕榈子药材中的主要成分,选用Hypersil BDS C18色谱柱(250 mm×4.6 mm,5μm),以甲醇(15%～0%)-乙腈(0%～15%)-0.3%磷酸水溶液(85%)为流动相进行梯度洗脱,洗脱时间90min,柱温为27℃,流速为0.800 ml/min,检测波长为220 nm,进样量为5μl。对9批不同来源的棕榈子进行指纹图谱相似度比较。结果:得到棕榈子药材的高效液相指纹图谱,各批次指纹图谱及对照图谱间相似度高。结论:该方法精密度、稳定性和重复性良好,可为棕榈子的质量控制提供参考。

Objective:To establish a HPLC fingerprint method for the similarities analysis of fructus trachycarpi from diffirent planting area.Method:The main components of fructus trachycarpi were extracted with 80% methanol by water bath reflux extraction method. Hypersil BDS C18 column(250 mm×4.6 mm,5μm)and Methanol(15%~0%)-acetonitrile(0%~15%)-0.3% phosphoric acid aqueous solution(85%)were used respectively as stationary phase and mobile phase in the chromatography system. The gradient elution time was 90 min,column temperature was 27 ℃,the flow rate was 0.800 ml/min,the detection wavelength was 220 nm,and the injection volume was 5 μl. The fingerprints similarity of 9 batches of fructus trachycarpi from different sources was compared.Results:The HPLC fingerprints of fructus trachycarpi were obtained,The similarities of fingerprints between each batch and the control were high.Conclusion:The method is precise,stable and reproducible,and can provide reference for quality control of fructus trachycarpi.
