摘要
目的考察白术提取物对耐甲氧西林金黄色葡萄球菌(MRSA)的作用。方法采用先加入过氧化氢水溶液氧化,再掺入抗坏血酸保护氧化产物方法提取白术有效成分。通过用牛津杯法测定其抗菌活性、二倍稀释法测定最低抑菌浓度(MIC),再将中耳炎模型成功的小鼠分为6组,分别为白术提取物高剂量组、中剂量组、低剂量组、氧氟沙星滴耳液组、万古霉素组和空白模型组。结果采用先氧化再保护氧化产物的方法得到白术提取物的收率为9.50%,白术提取物的MIC为0.781 mg/ml。与空白模型组比较,各实验组在给药1天后,菌落数量急速下降;各种浓度的白术提取物组在给药的第2天,菌落数量也明显下降。虽然与氧氟沙星和万古霉素相比,白术提取物起效比较慢,但在动物体内实验中对MRSA有效,并且不同浓度的白术提取物对MRSA控制效果也有差异,浓度越高,抑菌作用越强。结论本方法获得白术提取物的方法简单易行,产率高,经过体内外实验证明,可以单独用于抗MRSA,并且可以推测白术提取物对MRSA抗菌作用为浓度依赖性的。
Objective To study the effect of atractylodes macrocephala Koidz extraction on Methicillin resistant staphylococcus aureus(MRSA).Methods First we add hydrogen peroxide aqueous to oxygenize the sample,and then add into ascorbic acid which protects the oxidative product to extract effective consituent of Actractylodes macrocephala Koidz.Measure its antibacterial activity and the minimum inhibitory concentration by using oxford cup method and double dilution method.Next,we divide mice with Otisis Media into six groups:high dose of Atractylodes group,medium dose of Atractylodes group,low dose of Actractylodes group,Oxfoxacin Ear Drops group,Vancomycin group and blank model group.Results The extracting method of oxidizing before preserving oxidative product leads to 9.5% of Actractylodes macrocephala Koidz extractive,with a minimum inhibitory concentration of 0.781 mg/ml.Comparing with the blank model group,the number of bacterial colonies has sharply declined after 1 day of medication.On the second day of medication treatment,in the groups with Atractylodes in various concentrations,an obvious decline of bacterial colonies were also observed.Although the effect of Atractylodes came slowly than Oxfoxacin and Vancomycin,but it is proved to be effective to MRSA within animal bodies.Besides,there are differences among various concentrations of Atractylodes,higher concentrations tend to have better antibacterial function.Conclusions The method of extracting Atractylodes in this research is easy and productive.Through experiments in vivo and vitro,it proves that it can be independently used in MRSA,and we can speculate that the effect of Atractylodes macrocephala Koidz extractive is concentration-based.
