摘要
目的探讨环介导等温扩增(LAMP)技术在大学生弓形虫感染情况调查中的应用评价。方法随机选择2015—2017年本院检验专业学生共247人,抽取1 ml静脉血,离心取上清,采用氯仿-异戊醇方法提取DNA作为反应模板,以弓形虫529 bp序列为基础设计引物,进行LAMP扩增,同时采用ELISA方法进行Ig G抗体检查,比较分析二者的检测结果。结果 247名学生的弓形虫检测中,采用LAMP方法检测的阳性率为4.45%,采用ELISA检测的阳性率为6.48%,其中9例LAMP和ELISA检测的Ig G均为阳性。结论 LAMP技术可与ELISA联合检测,为临床弓形虫病的早期诊断提供了研究思路。
Objective To evaluate the application of loop-mediated isothermal amplification(LAMP) in the investigation of toxoplasma infection in college students.Methods 247 students majored in medical laboratory were selected randomly from our school during 2015 and 2017,1 ml venous blood were extracted,supernatant were collected,extract DNA template with chloroform-isoamyl alcohol method,primers were designed based on the toxoplasma 529 bp sequence,LAMP amplification were carried out,at the same time,Ig G antibody were tested by ELISA method,then the comparative analysis of the two test results were carried out.Results The positive rate of LAMP was 4.45%,and the positive rate was 6.48% by ELISA,9 cases of students were confirmed Ig G positive both by LAMP and ELISA.Conclusions The combination use of LAMP and ELISA provide a research idea for the early diagnosis of toxoplasmosis.
