离子色谱法同时测定阿拉伯胶水解后的单糖和糖醛酸 被引量:3

Determination of Monosaccharide and Uronic Acid after Hydrolysis of Acacia by Ion Chromatography

摘要 目的采用离子色谱法同时测定阿拉伯胶经酸水解后的单糖和糖醛酸含量。方法采用CarboPac PA20色谱柱,流动相为氢氧化钠溶液和醋酸钠溶液,梯度洗脱的方式,流速为0.5 m L·min^(-1),柱温为30℃,积分安培检测器。结果鼠李糖、阿拉伯糖、半乳糖、D-葡萄糖醛酸在0.5~10μg·m L^(-1)范围内与峰面积均呈良好的线性关系(r分别为0.999 3、0.999 4、0.999 5、0.999 8),平均加样回收率分别为99.1%(RSD=1.4%,n=6)、100.3%(RSD=1.1%,n=6)、98.1%(RSD=1.5%,n=6)、98.2%(RSD=1.2%,n=6)。结论本法操作简便、灵敏度高、结果准确可靠,可用于阿拉伯胶中单糖和糖醛酸的含量测定。 OBJECTIVE To establish an IC method for the determination of monosaccharide and uronic acid after acid hydrolysis of Acacia.METHODS The samples were separated on a CarboPac PA20 column ;the column was kept at 30℃,with the mobile phase consisting of Sodium hydroxide solution and sodium acetate solution in the gradient elution,at a flow rate of 0.5mL·min^-1.The elution was monitored at Integrated ampere detection detector.RESULTS There were good linears relationship between the rhamnose,Arabia sugar,galactose and D-glucuronic acid in the range of 0.5-10μg·mL^-1(r =0.9993,0.9994,0.9995,0.9998respectively).The average recovery rate was 99.1%(RSD =1.4%,n=6),100.3%(RSD =1.1%,n=6),98.1%(RSD =1.5%,n=6)and 98.2%, (RSD=1.2%,n=6)respectively.CONCLUSION The method is simple,sensitive,accurate and reliable,which can be used for the determination of monosaccharide and uronic acid in Acacia.
