少阴固本调枢法对STZ诱导糖尿病大鼠血清SOD、MDA、GSH-PX的影响

Effect of Shaoyin Kidney Supporting and Pivot Adjusting Method on Serum SOD,MDA,GSH-PX in STZ-induced Diabetic Rats

目的:观察少阴固本调枢法对STZ诱导的糖尿病大鼠血清SOD(超氧化物歧化酶)、MDA(丙二醛)、GSH-PX(谷胱甘肽过氧化物酶)含量的影响。方法:高脂高糖饲料+STZ诱导糖尿病模型,将造模成功的28只大鼠随机分为4组:模型组、少阴固本调枢法原方比例组、少阴固本调枢法经验用药比例组及吡格列酮组,每组7只;另取正常大鼠7只作对照。正常组和模型组灌服生理盐水,少阴固本调枢法原方比例组、少阴固本调枢法经验用药比例组分别按3 g/(kg·d)剂量灌胃,阳性对照组灌服吡咯列酮10mg/(kg·d),连续4周。观察大鼠体质量、血糖的变化,心包外抽血,离心取血清,ELISA检测大鼠血清INS、SOD、MDA、GSH-PX含量。结果:吡格列酮及少阴固本调枢法干预后,大鼠体质量较模型组明显减轻(P﹤0. 05),FPG与2 hPBG有不同程度的降低,并且血清胰岛素水平显著降低(P﹤0. 05);大鼠血清SOD、GSH-PX活性较模型组明显增强,MDA含量明显减少(P﹤0. 05);而少阴固本调枢原方比例组与经验用方比例组各项检测结果无明显差异(P> 0. 05)。结论:少阴固本调枢法具有降低2型糖尿病大鼠体质量、血糖及胰岛素水平,改善胰岛素抵抗,其作用机制可能与其抗氧化应激反应有关。

Objective :To observe the Shaoyin Kidney Supporting and Pivot Adjusting Method on serum SOD (superoxide dismutase),MDA (MDA),GSH -PX (glutathione peroxidase)in STZ -induced diabetic rats. Methods :28male STZ -induced diabetic Wistar rats were randomly divided into five groups :model group,the original prescnption of Shaoyin Kidney Supporting and Pivot Adjusting Method group and its clinical experience prescription group,positive control group with each 7 rats ,7SPF male Wistar rats were used as normal control. Normal group and model group were given normal saline,the Shaoyin Kidney Supporting and Pivot Adjusting Method original and cIinical experience prescription groups were given the corresponding Chinese medicine oral- ly,the positive control group was given pioglitazone,once a day for 4weeks,rats'weight was weighed and blood sugar was determined,serum INS,SOD,MDA and GSH -Px were determined by ELISA.Results:The wbight of rats in every medicating group was lightened,FBG and 2hPBG levels were lower at.different degree,serum INS was decreased significantly ,serum SOD and GSH -Px activities were enhanced and iDA content was reduced, there were significant differences between model and each medicating group (P <0.05);there was no obvious differences between the original and clinical experience prescription groups.Conclusion:Shaoyin Kidney Sup- porting and Pivot Adjusting Method can decrease weight,blood glucose and insulin levels,that is related to its anti -oxidative stress function.
