利湿化瘀法对湿瘀型慢性盆腔炎大鼠子宫组织Ⅰ型胶原纤维及IkKβ影响的研究 被引量:2

摘要 目的:通过观察利湿化瘀法对湿瘀型慢性盆腔炎大鼠子宫组织Ⅰ型胶原纤维及IκKβ蛋白表达的影响,进一步阐明利湿化瘀法的作用机制。方法:大鼠随机分为正常组和造模组;造模组沿子宫分叉处分别向双侧卵巢方向注入25%苯酚胶浆0. 04 mL,再随机分成模型组,中药高、低剂量组和妇乐组。正常组和模型组给予生理盐水灌胃,其余组分别给以相应剂量的利湿化瘀中药和妇乐颗粒混悬液灌胃。21 d后,腹腔麻醉下取子宫组织,采用Masson法检测Ⅰ型胶原纤维; Western Blot法检测IκKβ(IKβ激酶)蛋白表达。结果:模型组大鼠子宫组织可见大量Ⅰ型胶原纤维,中药低剂量组胶原纤维量表达少于模型组,但高于中药高剂量组和妇乐组,中药高剂量组和妇乐组胶原纤维量表达接近正常组,无明显区别。与模型组比较,中药高剂量组、低剂量组、妇乐组均能降低IκKβ蛋白的表达,差异有统计学意义(P <0. 05);中药高剂量组和妇乐组比较,差异不显著(P <0. 05)。结论:利湿化瘀法通过干预大鼠子宫组织Ⅰ型胶原纤维及IκKβ蛋白的表达,促进细胞凋亡、抑制组织粘连及纤维化的形成,从而对湿瘀型慢性盆腔炎具有良好临床疗效。 Objective:To observe the effects of Removing Dampness and Stasis Method on the expressions of collagen type I fibers and IKK^protein in uterus of rats with chronic pelvic inflammatory disease of dampness -stasis type,and further explain the mechanism of Removing Dampness and Stasis Method.Methods:Rats were randomly divided into two groups:normal group and model group;model group was injected 25% phenol glue 0.04mL into bilateral ovaries along the uterine bifurcation,and then randomly divided into model group, high and low dose group of Chinese medicine and Fule positive group.Normal group and model group were given saline by gastric lavage,and the other groups were given corresponding doses of Chinese herbal medicine for re- moving dampness and stasis and Fule granule suspension respectively.Twenty -one days later,uterine tissues were taken under abdominal anesthesia,and type I collagen fibers were detected by Masson's method,and the expression of type I IκKβ(IKβ kinase)protein was detected by Western Blot method.Results:A large number of collagen type I fibers were found in the uterus of rats in the model group.The amount of collagen fibers in the low dose group was less than that in the model group,but higher than that in the high dose group and Fule group.Compared with model group,the expression of IκKβ protein in high dose group,low dose group and Fule group were decreased,the difference was significant (P <0.05),but there was no significant difference between high dose group and Fule group (P <0.05).Conclusion:Removing Dampness and Stasis Method can promote apoptosis,inhibit the formation of tissue adhesion and fibrosis by interfering with the expression of collagen type I fibers and IκKβ protein in rat's uterus,so as to effectively treat chronic pelvic inflammatory disease of damp - stasis type.
