摘要
目的:观察膝骨关节炎(KOA)模型大鼠鹤顶、委中在腧穴敏化过程中局部穴区肥大细胞(MC)超微结构改变,探讨腧穴敏化的MC机制。方法:将18只SD大鼠随机分为正常组、生理盐水组和模型组,每组6只。模型组采用单碘乙酸盐膝关节注射的方法制备大鼠KOA模型。造模后14天采用酶联免疫吸附(ELISA)法检测大鼠血清IL-1β、CTX-Ⅱ水平;取鹤顶、委中穴区皮肤和皮下结缔组织,采用透射电子显微镜观察MC超微结构改变。结果:透射电子显微镜下观察到模型组鹤顶穴穴区MC周围有颗粒脱出,颗粒膜相互融合,脱颗粒后遗留有空泡;ELISA法结果显示模型组大鼠血清IL-1β、CTX-Ⅱ水平均显著高于正常组与生理盐水组(P <0. 01);模型组鹤顶穴穴区MC胞内致密颗粒占比较正常组及生理盐水组均明显降低(P <0. 01);模型组委中穴穴区MC胞内致密颗粒占比与正常组及生理盐水组相比较差异无统计学意义(P> 0. 05)。结论:MC参与了与疾病相关的腧穴敏化过程,特定疾病状态下的腧穴敏化具有腧穴特异性。
Objective:To observe the ultrastructural changes of mastocytes around sensitized acupoints in ratswith knee osteoarthritis(KOA),and to explore the mastocytes mechanism of acupoint sensitization. Methods:18SD rats were randomly divided into the normal group,the normal saline group,and the model group,with 6 ratsin each group. Mono - iodoacetate was injected into the knee joints to establish KOA model. Serum levels ofIL -1β and CTX - Ⅱ were tested by ELISA method after 14 days of modeling. Ultrastructural changes of mas-tocyte in the skin and subcutaneous connective tissue around Heding(EX - LE2) and Weizhong(BL40) wereobserved under transmission electron microscope. Results:Transmission electron microscope showed that therewere granular prolapse and granular membrane fusion around the mastocytes,and some vacuoles were seen afterdegranulation in the model group around EX - LE2. The serum levels of IL -1β and CTX - Ⅱwere significantlyhigher in the model group than those in the normal group and the normal saline group(P <0. 01). In terms ofthe percentage of dense granules in mastocytes,it was significantly decreased in the model group than those inthe normal group and the normal saline group around EX - LE2(P <0. 01); however,there was no statisticaldifference among the three groups around BL40(P >0. 05). Conclusion:Mastocytes participate in the process ofKOA - induced acupoint sensitization. There was acupoint specificity in acupoint sensitization for some specificdisease.
