醒髓汤对急性上颈脊髓损伤模型大鼠四肢运动功能及脊髓组织DAF表达影响 被引量:2

摘要 目的:分析醒髓汤对急性上颈脊髓损伤模型大鼠四肢运动功能及脊髓组织血小板活化因子(PAF)表达影响。方法:选取SPF级Wistar雄性大鼠40只,制备急性脊柱损伤模型,随机数字表法将其分成两组,观察组(20只)与对照组(20只),术后观察组大鼠灌胃醒髓汤,每天两次,每次2ml;对照组灌胃等剂量生理盐水,每天两次,每次2ml,两组大鼠均连续灌胃14天;使用BBB分级法检测给药1、3、7、14天时大鼠四肢功能状况,免疫组化法检测大鼠受损组织内Caspase-3表达的阳性细胞,检测免疫阳性产物积分光密度数值(IOD),ELISA法检测大鼠脊髓组织内PAF含量状况。结果:给药3天、7、14天时观察组大鼠BBB评分均高于对照组,差异有统计学意义(P<0.05);给药1、3、7、14天观察组大鼠Caspase-3阳性细胞IOD数值均低于对照组,差异有统计学意义(P<0.05);给药1、3、7、14天观察组大鼠随机组织内PAF表达量均低于对照组,差异有统计学意义(P<0.05)。结论:醒髓汤可显著降低急性上颈脊髓受损模型大鼠脊髓组织内PAF含量,改善其四肢运动功能。 Objective: To analyze the effect of Xingjie Decoction on the motor function of extremities and the expression of platelet activating factor( PAF) in spinal cord tissue for rats with acute upper cervical spinal cord injury. Methods: Forty male Sprague-Dawley Wistar rats were selected to prepare acute spinal injury models. The randomized digital table method was used to divide them into two groups. The observation group( 20 rats) and the controlled group( 20 rats). The decoction was decocted twice a day,2 ml each time; The controlled group was given an equal dose of normal saline twice a day,2 ml each time,and the rats of both groups were continuously gavaged for 14 days; BBB grading was used to detect the functional status of rats’ limbs was measured on day 1,day 3,day 7 and day 14. The positive cells of Caspase-3 in injured tissues were detected by immunohistochemistry. The integrated optical density( IOD) of immunopositive products was detected. The ELISA was used to examine the spinal cord of rats. PAF content status. Results: The BBB scores of the observation group were higher than those of the controlled group on the 3 rd,7 th,and 14 th day after administration( P < 0. 05); Caspase-treated rats were observed on the 1,3,7 and 14 days. The IOD value of positive cells was lower than that of the control group,and the difference was statistically significant( P <0. 05). The expression of PAF in the random tissue of the observation group rats on the 1,3,7 and 14 days after administration was lower than that of the controlled group,with statistical significance( P < 0. 05). Conclusion: Xingjie Decoction can significantly reduce the content of PAF in the spinal cord of rats with acute upper cervical spinal cord injury and improve their motor function.
