当归拈痛汤对急性痛风性关节炎大鼠关节肿胀及组织炎性因子影响

Effect of Danggui Niantong Decoction on Joint Swelling and Tissue Inflammatory Factors for Rats with Acute Gouty Arthritis

目的:分析当归拈痛汤对急性痛风性关节炎(AGA)大鼠关节肿胀及组织炎性因子影响。方法:选取SD雄性大鼠22只,依据随机数字表法将其分成两组,对照组(11只)与观察组(11只),使用250mg/kg/d乙胺丁醇、200mg/kg/d腺嘌呤灌胃制备高尿酸血症(HUA)模型,而后在大鼠右踝关节外侧后边,注入尿酸钠溶液0. 2ml制备AGA模型,对照组使用蒸馏水灌胃口,观察组当归拈痛汤灌胃,观察大鼠造模及给药后状态变化,造模第1天及给药后7天大鼠体重与尿量状况,给药1、2、3天分别测量大鼠关节肿胀情况,计算肾脏脏器指数,检测血清内血尿酸含量状况,免疫组化法检测大鼠滑膜组织COX-2、TNF-α及IL-1β含量状况。结果:给药后7天,观察组大鼠体重高于对照组,尿量低于对照组,差异有统计学意义(P<0. 05);给药后观察组大鼠关节肿胀改善程度优于对照组,差异有统计学意义(P<0. 05);观察组大鼠血尿酸值,肾脏脏器指数,滑膜组织内COX-2、IL-1β及TNF-α含量均低于对照组,差异有统计学意义(P<0. 05)。结论:当归拈痛汤可明显改善AGA大鼠关节肿胀程度,降低滑膜组织内炎症因子含量。

Objective: To analyze the effect of Danggui Niantong Decoction on joint swelling and tissue inflammatory factors for rats with acute gouty arthritis( AGA). Methods: Twenty-two male Sprague-Dawley rats were divided into two groups according to the random number table method,The controlled group( n = 11) and the observation group( n = 11) received 250 mg/kg ethambutol and 200 mg/d adenine gavage to prepare hyperuricemia( HUA) model,and then in the rat right ankle joint outside the lateral injection of sodium urate solution 0. 2 ml preparation AGA model,the controlled group used distilled water from mouth,the observation group was given Danggui Niantong Decoction. The rats were observed after modeling and the change of state,the first day of modeling and 7 days after the administration of body weight and urine output of rats,administered 1,2,3 days were measured in rats joint swelling,kidney contents of serum uric acid in serum were detected by immunohistochemical method. The levels of COX-2,TNF-α and IL-1β were detected by immunohistochemistry. Results: Seven days after the administration,the body weight of the observation group was higher than that of the controlled group,and the urine output was lower than that of the controlled group( P < 0. 05). After treatment,the degree of joint swelling in the observation group was better than that of the controlled group. The difference was statistically significant( P < 0. 05); The content of serum uric acid,index of kidney,COX-2,IL-1β and TNF-αin synovial tissue in the observation group were lower than those in the controlled group significance( P <0. 05). Conclusion: Danggui Niandong Decoction can significantly improve the degree of joint swelling in AGA rats and reduce the content of inflammatory cytokines in synovial tissue.
