摘要
目的探讨济宁地区健康成年人外周血淋巴细胞亚群的变化特点,建立济宁地区健康成年人淋巴细胞亚群参考区间。方法使用美国BD Multitest?IMK四色特异性淋巴细胞亚群荧光抗体,采用BD公司FACSCalibur流式细胞仪进行检测。对2015年12月至2017年6月至济宁医学院附属医院以及济宁市第一人民医院体检的共1 800例健康成人外周血淋巴细胞亚群进行分析。采用SPSS21.0软件统计分析结果。结果男性组的NK细胞比例为(13.89±7.02)%,明显高于女性组的(12.73±6.43)%,CD4/8双阴性T淋巴细胞为(2.91±1.55)%,明显低于女性组的(3.23±1.71)%,差异均有统计学意义(P<0.05)。济宁地区健康成年人外周血淋巴细胞参考值T淋巴细胞(CD3+)为53.10%~82.45%,B淋巴细胞(CD3-CD19+)为5.17%~17.64%,T辅助细胞(CD3+CD4+)为35.92%~9.17%,T抑制细胞(CD3+CD8+)为16.17%~42.58%,NK细胞(CD3-CD16+CD56+)为4.61%~26.27%,NKT细胞(CD3+CD16+CD56+)为1.07%~10.58%,双阳性T细胞(CD3+CD4+CD8+)为0.08%~2.71%,双阴性T细胞(CD3+CD4-CD8-)为0.53%~6.51%。结论一个地区可以建立健康成年人外周血淋巴细胞亚群参考区间,为机体免疫状态评估、研究、临床疾病诊断及预后评估提供参考依据。
Objective To analyze the changes of peripheral blood lymphocyte subsets among healthy adults in Jining area, and to establish reference value of peripheral blood lymphocyte subsets. Methods A total of 1 800 healthy adults from Affiliated Hospital of Jining Medical University and Jining No.1 People’s Hospital from Dec. 2015 to Jun. 2017 were enrolled in the research. BD Multitest? IMK kit (four-color specific lymphocyte subset fluorescent antibody) and BD FACScalibur flow cytometry were used for the lymphocyte subsets. The results were analyzed using SPSS21.0 software. Results The percentages of NK lymphocytes in males were (13.89±7.02)%, significantly higher than (12.73± 6.43)% in females (P<0.05), while the percentages of CD4- CD8- T cells in males were (2.91±1.55)%, significantly lower than (3.23 ± 1.71)% in the control group (P<0.05). The reference of peripheral blood lymphocyte subsets among healthy adults in Jining area was as follows: 53.10%-82.45% for T lymphocytes (CD3+), 5.17%-17.64% for B lymphocytes (CD3-CD19+), 35.92%-9.17% for T help lymphocytes (CD3+ CD4+), 16.17%-42.58% for T suppressor cells (CD3+ CD8+), 4.61%-26.27% for NK lymphocytes (CD3- CD16+ CD56+), 1.07%-10.58% for NKT lymphocytes (CD3+ CD16+ CD56+), 0.08%-2.71% for CD4/8 double positive T cells (CD3+ CD4+ CD8+), 0.53%-6.51% for CD4/8 double negative T cells (CD3+ CD4- CD8-). Conclusion The reference ranges of peripheral blood lymphocyte subsets among healthy adults could be established, to provide reference for the evaluation and research of immune status, as well as the diagnosis and prognosis evaluation of clinical diseases.
