Quality evaluation and cluster analysis of Pinellia ternata from different origins based on HPLC fingerprints

摘要 目的基于指纹图谱与聚类分析方法对不同产地半夏进行质量评价,以期对半夏药材的产地选择及质量控制提供参考。方法利用HPLC法对半夏药材进行质量分析,所获数据用中药色谱指纹图谱相似度评价系统构建半夏药材的指纹图谱,并应用SPSS 22.0统计软件进行系统聚类分析,以共有色谱峰的峰面积为变量建立树状图,分析产地因素对药材的影响。色谱柱为Waters HSS T3 C18(4.6 mm×250mm,5μm)色谱柱;流动相为乙腈-水溶液,梯度洗脱;检测波长:260 nm;流速:1.0 mL·min-1;柱温:25℃;进样量:20μL。结果半夏药材HPLC指纹图谱的12个峰确定为共有峰,其中确定10批半夏药材的相似度为0.906~0.973,10批半夏具有较好的一致性,稳定性较好。聚类分析结果表明,半夏药材从质量上可分为两大类,分类受地域影响较大。结论本方法快捷简便、真实可靠,可为半夏药材的产地选择及质量控制提供参考。 Objective To evaluate the quality ofPinellia ternata from different habitats based on fingerprints and cluster analysis,and to provide reference for the selection and quality control ofPinellia ternata.Methods The quality ofPinellia ternata was analyzed by HPLC.The obtained data were constructed with the Chinese medicine chromatographic fingerprint similarity evaluation system.SPSS 22.0statistical software was used for systematic clustering analysis.A tree diagram was established with the peak area of the common chromatographic peak as a variable.The chromatographic column was Waters HSS T3C18(4.6mm×250mm,5μm).The flow phase was acetonitrile-aqueous solution gradient elution.The detection wavelength was 260nm;the flow rate was 1.0 mL·min^-1;the column temperature was 25℃ ;and the sample quantity was 20μL.Results The 12peaks of HPLC fingerprints ofPinellia ternata were identified as common peaks,and the similarity of 10batches of Pfllellia ternata ranged 0.906to 0.973.The 10batches ofPinellia ternata had better consistency and better stability.Cluster analysis showed that the medicinal herbs ofPinellia ternata were divided into two categories in quality,and the classification was greatly affected by the region.Conclusion This method is fast,simple,and reliable,which can be used as a reference for the selection of origin and quality control ofPinellia ternata.
