HPLC测定肉苁蓉配方颗粒的含量及指纹图谱研究

Study on the fingerprints and content determination of Cistanches herba dispensing granules by HPLC

目的采用HPLC法测定肉苁蓉配方颗粒的含量,并研究其指纹图谱。方法采用HPLC法测定肉苁蓉配方颗粒中松果菊苷和毛蕊花糖苷的含量,并建立肉苁蓉配方颗粒的指纹图谱,同时利用相似度评价软件比较分析10批肉苁蓉配方颗粒的色谱图。结果各批次肉苁蓉配方颗粒指纹图谱的相似度良好,且具有良好的稳定性和特征性;26.8389~429.4220mg·L-1松果菊苷与峰面积呈良好的线性关系(r^2=0.9999),平均加样回收率为97.34%(RSD=0.76%,n=9);5.2817~84.5069mg·L^-1毛蕊花糖苷与峰面积呈良好的线性关系(r2=0.9999),平均加样回收率为95.55%(RSD=0.17%,n=9)。结论所用肉苁蓉配方颗粒指纹图谱及含量测定的方法简便、准确,可为肉苁蓉配方颗粒的质量控制提供依据。

OBJECTIVE To determine the contents of the active ingredients and the fingerprint chromatogram of Cistanches herba dispensing granules( CHDG) using HPLC methods. METHODS High performance liquid chromatography( HPLC) was applied for the content determination of echinacoside and acteoside in CHDG as well as the establishment of HPLC fingerprints. The similarity of 10 batches of samples were evaluated by using the similarity evaluation software. RESULTS The fingerprints of CHDG were similar in all batches and had good stability and characteristics. Echinacoside showed a good linear relationship( r2= 0. 9999) at 26. 8389-429. 4220 mg·L-1 with average recovery of 97. 34%( RSD = 0. 76%,n = 9). Acteoside showed a good linear relationship( r2= 0. 9999) at5. 2817-84. 5069 mg·L-1 with average recovery of 95. 55%( RSD = 0. 17%,n = 9). CONCLUSION The method is easy to operate and accurate,and could be used for the quality control of CHDG.
