摘要
目的建立一种快速、可靠、灵敏检测植物油中维生素E含量的方法。方法采用50%KOH-Et OH溶液中加入10%抗坏血酸作抗氧化剂,在80℃水浴中加热0.5 h,对植物油中的维生素E进行皂化处理,制备供试品溶液;采用Acquity UPLC C8色谱柱(100 mm×2. 1 mm,1. 7μm),流动相A为水、B为甲醇,梯度洗脱,流速0. 25 m L·min-1,检测波长285 nm。结果2. 19~203. 5μg·m L-1维生素E与峰面积的线性相关系良好(Y=726. 55X-130. 46,r2=0. 9996),检测限和定量限分别为10.7、26.9 ng·m L-1,平均回收率为82. 75%,重复性的RSD=3. 60%,耐用性良好,表明建立的方法能够满足植物油中维生素E的测定要求。在13种植物油中均检出维生素E,其中,小麦胚芽油中维生素E的含量最高,沙棘油次之。结论所用方法可快速、灵敏、精确地测定13种植物油中维生素E的含量。
OBJECTIVE To establish a fast,reliable,and sensitive method for the determination of Vitamin E( Vit E) in vegetable oils. METHODS The Vit E in vegetable oil was saponified by 50% KOH-Et OH solution added with 10% ascorbic acid as an antioxidant,heating in a water bath at 80 ℃ for 0. 5 h,to prepare sample solution. The chromatographic conditions were as follows:UPLC C8 column of Acquity( 100 mm × 2. 1 mm,1. 7 μm). The mobile phase A was water,the mobile phase B was methanol. The gradient elution was performed at a flow rate of 0. 25 m L·min-1,with the detection wavelength set at 285 nm. RESULTS The methodology study showed that the Vit E’s concentration was in a linearity relationship within the range of 2. 19-203. 5 μg·m L-1( Y = 726. 55 X-130. 46,r2= 0. 9996). The detection limit was 10. 7 ng,and the quantification limit was 26. 9 ng·m L-1. The average recovery was 82. 75%,and the RSD of repeatability was 3. 60%. The durability indicated that the method can determine Vit E’s distribution in vegetable oil. Vit E was detected in 13 kinds of edible vegetable oil. The highest content of Vit E was found in wheat germ oil followed by the seabuckthorn oil. CONCLUSION The method is rapid,sensitive and accurate,suitable for determination of Vit E in different vegetable oils.
