碳酸酐酶Ⅻ在口腔鳞癌中的表达及意义

摘要 目的探讨碳酸酐酶Ⅻ(carbonic anhydraseⅫ,CAⅫ)在口腔鳞癌(oral squamous cell carcinoma,OSCC)中的表达及其与临床病理特征的关系。方法采用免疫组织化学的方法检测CAⅫ在60例OSCC组织及20例正常口腔黏膜组织中的表达,酶联免疫法(ELISA)检测CAⅫ在30例OSCC患者及10例健康人血清中的表达,观察其与OSCC的相关性。结果 60例OSCC组织中有43例CAⅫ呈阳性表达,阳性率为71%,明显比正常口腔黏膜组织高(10%)(P <0. 05); CAⅫ的组织表达水平和OSCC的临床分期以及肿瘤直径显著相关(P <0. 05),但是和患者的年龄、性别、淋巴结转移以及OSCC的分化程度无关(P> 0. 05)。OSCC患者血清中CAⅫ的表达量显著高于健康对照者(Z=4. 685,P=0. 00),但在OSCC的各项临床病理特征之间差异无统计学意义(P> 0. 05)。结论 CAⅫ可能在OSCC的生长、侵袭中发挥着重要作用;定量检测血清CAⅫ在OSCC的诊疗中具有一定的应用价值。 Objective To explore the expression of CAⅫ and its correlation with clinical pathologic findings in the oral squamous cell carcinoma(OSCC).Methods Immunohistochemistry method was applied to measure the expression of CAⅫ in 60 cases of OSCC and 20 cases of normal oral mucosa tissues,serum CAⅫ was measured with ELISA in 30 patients with OSCC and 10 healthy controls.The relationship of CAⅫ with the clinicopathologic features of OSCC patients were analyzed.Results CAⅫ was positive in 43/60 cases of OSCC,with the positive rate of 71%,which was significantly higher than that of the normal oral mucosa(P <0.05).The expression of CA Ⅻ was siginificantly associated with the clinical stages,tumor size of OSCC (P <0.05),but was not associated with sex,age,lymph node metastasis of patients with OSCC and differentiation of OSCC (P >0.05).Serum levels of CAⅫ were higher in the patients with OSCC than heathy controls (Z=4.685,P =0.00),there was no siginificant difference among clinicopathologicl characters (P >0.05).Conclusion CAⅫ might play an important role in the development and invasion of OSCC.Quantification of serum CAⅫ may be a valuable tool in the diagnosis and treatment of OSCC.
