右归丸对大鼠骨髓间充质干细胞中TGF-β、BMP-7、BGP表达影响实验研究

Experimental study on the effect of Yougui Pill on the Expression of TGF-β,BMP-7 and BGP in Rat Bone Marrow Mesenchymal Stem Cells

目的:通过对大鼠骨髓间充质干细胞中TGF-β、BMP-7、BGP蛋白表达的分析,探究右归丸含药血清通过TGF—β信号通路诱导BMSCs向成骨细胞分化的机制,从而为“肾主骨”这一理论提供实验依据。方法:42只SPF级SD雌性大鼠,体质量(210±20)g,随机分为:正常组、诱导液组、右归丸组、福善美组、骨疏康组各8只,对大鼠灌胃5d,制备含药血清,并分离和培养大鼠BMSCs,观察不同实验组含药血清对成骨细胞中TGF—β、BMP-7、BGP蛋白表达的影响。结果:与正常组相比,其余各实验组均不同程度上调了TGF—β、BMP-7、BGP的表达水平,并且右归丸组与其余各组相比,结果有统计学差异(P<0.01)。结论:①正常大鼠BMSCs中存在TGF—β、BMP-7、BGP蛋白表达;②右归丸含药血清能够通过调控TGF—β、BMP-7、BGP的蛋白浓度促进BMSCs向成骨细胞分化,以起到对骨质疏松症的防治作用。

Objective:To investigate the mechanism of differentiation of BMSCs into osteoblasts induced by Yougui Pill containing serum through TGF-β signaling pathway by analyzing the protein expression of TGF-β,BMP-7 and BGP in rat bone marrow mesenchymal stem cells,so as to provide experimental basis for the theory of "kidney storing essence".Methods :The 42 SPF SD rats were randomly divided into:normal group,induced liquid group,Yougui Pill group,Fosamax group and Gushukang group,each group had 8 rats,then the rats were administered orally to the rats for 5days.Prepared drug contained serum,BMSCs of rats were isolated and cultured.The effects of drug-containing serum on the expression of TGF-β,BMP-7and BGP protein in osteoblasts of different experimental groups were observed.Results : Compared with the normal group,the expression levels of TGF-β,BMP-7 and BGP were increased in different experimental groups,the expression level of the protein increased significantly in Yougui Pill group,the results were statistically different (P<0.01).Conclusion :(1)The protein expressions of TGF-β,BMP-7and BGP can be found in normal rat bone tissue;(2)The contents of TGF-β ,BMP-7 and BGP in Yougui Pill group were significantly higher than those in normal group,which indicated that Yougui Pill containing serum could regulate the protein concentration of TGF-β,BMP-7 and BGP and promote the proliferation of bone marrow mesenchymal stem cells differentiating into osteoplast,and thus play a role in prevention and treatment of osteoporosis.
