葫芦素D对血管新生及bFGF诱导PLCγ/Erk1/2通路激活影响 被引量:1

Anti-angiogenesis Effect of Cucurbitacin D by Inducing bFGF-Activated PLCγ/Erk1/2 Pathway

摘要 目的:评价葫芦素D对血管内皮细胞增殖、迁移、成管作用的影响及其作用机制。方法:采用人脐静脉血管内皮细胞株(EA.hy926),通过CCK8细胞增殖检测、划痕实验、管腔形成实验检测葫芦素D对血管新生各环节的作用,并通过Western Blot法检测葫芦素D对bFGF介导的细胞信号通路激活的抑制作用。结果:葫芦素D对血管内皮细胞活性影响较低,同时能够明显抑制血管内皮细胞的迁移、成管能力;能够抑制bFGF刺激下FGFR2及其下游信号通路关键蛋白PLCγ、Erk1/2的磷酸化。结论:葫芦素D具有抑制血管新生的作用,其机制可能与抑制bFGF介导的信号通路激活有关。 Objective:To evaluate the effect of cucurbitacin D on the proliferation,migration and tube formation of vascular endothelial cells and its mechanism of action. Methods:The human umbilical vein endothelial cell line(EA.hy926)was used to detect the effects of cucurbitacin D on angiogenesis by CCK8 cell proliferation assay,scratch test,and lumen formation assay. Inhibition of bFGF-mediated activation of cell signaling pathways of cucurbitacin D was detected by Western Blot assay. Results:Cucurbitacin D had a low effect on the activity of vascular endothelial cells,and it could also significantly inhibit the migration and tube formation of vascular endothelial cells. It also inhibited the phosphorylation of FGFR2 and its key downstream proteins PLCγ and Erk1/2 under the stimulation of bFGF. Conclusion:Cucurbitacin D inhibits angiogenesis and its mechanism may be related to the inhibition of bFGF-mediated signal pathway activation.
