基于《中医方剂大词典》活血化瘀方治疗心系疾病用药规律数据挖掘研究

Data Mining Analysis of Regularity of Formulae for Promoting Blood Circulation to Remove Stasis for the Cardiovascular Disease

目的:基于中医传承辅助平台系统(V2.5)软件,对《中医方剂大辞典》中活血化瘀方剂在治疗心系疾病方面的用药特点进行探讨并分析其规律,以期对活血化瘀法治疗心系疾病进一步完善补充。方法:利用《中医方剂大辞典》系统搜集用于治疗心系疾病的具有活血化瘀功效的方剂,应用“中医传承辅助平台”系统的数据挖掘方法,如频数统计、复杂网络分析、关联规则分析、熵方法分析等,分析药物频数、关联规则、处方规律、核心药物组合、新方组合等用药及组方规律。结果:最终纳入方剂327首,涉及药物275味;使用频率最高的中药为当归(158次,48.31%)、川芎(100次,30.58%)、赤芍(89次,27.21%);最常见的对药为川芎-当归(120次,50.84%),其次为赤芍-当归(104次,44.06%);最常见的角药为赤芍-当归-肉桂(49次,20.76%),赤芍-川芎-当归(49次,20.76%);基于关联规则发现的常见药物组合:赤芍→当归(置信度1),川芎+白芍→当归(置信度1),延胡索+川芎→当归(置信度1),基于复杂网络发现的核心药物有当归、川芎、赤芍、人参、肉桂、生地黄;通过熵方法提取出来的新方组合12个。结论:本研究进一步的挖掘了活血化瘀方在历史过程中治疗心系疾病的核心用药及组方规律,其中当归、川芎、赤芍等核心药物常配伍温阳药、益气药、理气药等,其治法多为活血化瘀,益气温阳,行气止痛,进而为活血化瘀方在心系疾病治疗上提供补充借鉴,以及为下一阶段活血化瘀方干预心血管疾病的新药研发方面提供补充参考。

Objective:Based on inheritance platform system,using date mining to explain regularity of formulae for promoting blood circulation to stasis for cardiovascular disease in the database of Dictionary of Chinese Medicine Prescription(DCMP). Methods:Selecting the formulae for promoting blood circulation to remove stasis for the cardiovascular disease,analyze the rules of the law of prescriptions core herbs and association rules by using rule analysis,improved mutual information law,complex system entropy clustering,unsupervised hierarchical clustering method. Results:Based on the analysis of 327 cases of prescriptions,including 275 kinds of herbs,in this book,the commonest herbs conclude Chinese angelica (158 cases,48.31%),rhizome of Chuanxiong(100 cases,30.58%),red peony(89 cases,27.21%); The most frequent couple combination respectively are rhizome of Chuanxiong-Chinese angelica(120 cases,50.84%),red peony-Chinese angelica(104 cases,44.06%);The commonest triple combination respectively are red peony-Chinese angelica-cinnamon(49 cases,20.76%)and red peony-rhizome of Chuanxiong-Chinese angelica(49 cases,20.76%);Based on the association rules,frequent medicine combination are red peony → Chinese angelica(CC=1),rhizome of Chuanxiong + white peony → Chinese angelica(CC=1),Rhizoma Corydalis + rhizome of Chuanxiong → Chinese angelica(CC=1). Based on complex network analysis,the core medicinals identified are Chinese angelica,rhizome of Chuanxiong,red peony,ginseng,cinnamon,dried rehamnnia root;The new compounds extracted by entropy method involved total 12. Conclusion:The research further explored the core medicinals and association rules of formulae for promoting blood circulation to remove stasis for the cardiovascular diseases. The core medicinals,like Chinese angelica,rhizome of Chuanxiong,red peony,often cooperate warming Yang herbs,Qi-invagorating herbs and Qi-regulating herbs,etc. The methods of therapy conclude promoting blood circulation to remove stasis,invigorating Qi and warming Yang and promoting Qi circulation to relieve pain. The research will provide reference for therapy of promoting blood circulation to remove stasis for the heart diseases,and provide guidance for the new drug research of formulae for promoting blood circulation to remove stasis in the area of cardiovascular disease next stage.
