摘要
为了获得白羊草(Bothriochloa ischaemum)转录组数据库和差异表达基因,通过Illumina HiSeq4000高通量测序技术对白羊草对照组和干旱胁迫组进行转录组测序。经组装分析共获得118580条Unigenes;将所得到的Unigenes与基因库数据进行比较,对Unigenes进行功能注释,共50070条Unigenes获得功能注释。白羊草转录组中的Unigenes根据GO功能大致可分为细胞组分、分子功能和生物学过程三大类共54个分支。通过将所有Unigenes与KOG数据库比对,共有1586条Unigenes可归为25类。以KEGG数据库作为参考,依据代谢途径可将Unigene定位到116个代谢途径分支。通过差异性分析发现,差异性表达基因共3987条,其中上调基因占39.45%,下调基因占60.55%。本研究为白羊草分子生物学研究提供了宝贵的基因组数据库资源。
In order to obtain the transcriptome database and differentially expressed genes of Bothriochloa ischaemum,transcriptome sequencing of the control group and drought stress group was performed with Illumina HiSeq 4000 high-throughput sequencing technology.A total of 118580 unigenes were obtained from the assembly analysis.All unigenes were compared with the gene database,and the functional annotations were performed on unigenes,about 50070 unigenes were got.According to the function of GO,unigenes in the transcriptome of B.ischaemum could be roughly divided into three categories:cellular component,molecular function and biological process,with a total of 54 branches.All unigenes were compared with the KOG database; there were 1586 unigenes which were classi fi ed to 25 terms.Using the KEGG database as a reference,unigenes could be mapped to 116 classes depending on the metabolic pathway.Through the differentially expressed analysis,we got 3987 differentially expressed genes,the up-regulated genes accounted for 39.45%,the down-regulated genes accounted for 60.55%.This study provides a valuable genome data source for molecular biology research of B.ischaemum.
