甜菜M14品系咖啡酰辅酶A-O-甲基转移酶提高植物抗逆性功能分析 被引量:5

摘要 为研究甜菜M14品系咖啡酰辅酶A-O-甲基转移酶基因(BvM14-CCoAOMT)的功能,本研究采用叶盘法将M14品系咖啡酰辅酶A-O-甲基转移酶基因转化烟草,获得T1代转基因植株,而后以野生型及转空载体植株为对照,分别在对照、100 mmol/L NaCl、200 mmol/L NaCl、100 mmol/L甘露醇及200 mmol/L甘露醇的培养条件下进行植株的抗逆性分析。研究发现在100、200 mmol/L NaCl盐胁迫条件下,转基因植株的鲜重、木质素含量以及叶绿素含量均显著高于对照植株。此外转基因植株在100 mmol/L甘露醇胁迫条件下根长和叶绿素含量显著高于对照植株,根长和叶绿素含量分别为0.68 cm和0.31 mg/(g·鲜重),而野生型植株的根长和叶绿素含量分别为0.51 cm和0.18 mg/(g·鲜重)。转M14品系咖啡酰辅酶A-O-甲基转移酶基因烟草植株对盐和干旱胁迫具有较强抗性,这为进一步深入研究M14品系咖啡酰辅酶A-O-甲基转移酶基因功能奠定重要基础。 To study the function of caffeoyl CoA O-methyltransferase gene (BvM14-CCoAOMT)in sugar beet M14 strain,BvM14-CCoAOMT was transformed into tobacco with leaf disc method,and the transgenic plants of T1 generation were obtained.Furthermore,compared with wild type and transformed empty carrier plants, the resistance of positive transgenic plants was analyzed under the conditions of control,100mmol/L NaC1, 200mmol/L NaCl,100mmol/L mannitol and 200mmol/L mannitol.The results showed that under 100mmol/L NaC1and 200mmol/L NaC1stress,the fresh weight,lignin content and chlorophyll content of the transgenic plants were significantly higher than those of control plants.In addition,the root length and chlorophyll content of transgenic plants under 100mmol/L mannitol were significantly higher than those of the control plants.The root length and chlorophyll content of transgenic plants was 0.68cm and 0.31mg/(g ·fresh weight), respectively,while the root length and chlorophyll content of wild type plants was 0.51cm and 0.18mg/(g· fresh weight),respectively.The transgenic tobacco plants with BvM14-CCoAOMT gene have strong resistance to salt and drought stress,and the study provides a theoretical basis for further study on BvM14-CCoAOMT function.
