柴胡疏肝散精简方对利血平致抑郁样模型大鼠行为学及中缝核内儿茶酚胺的影响 被引量:7

摘要 目的:观察柴胡疏肝散精简方对利血平致抑郁样模型大鼠行为学及中缝核内儿茶酚胺的影响,明确柴胡疏肝散精简方的抗抑郁作用,探究柴胡疏肝散精简方的抗抑郁机制。方法:将48只大鼠随机分为4组,每组12只,分别为空白组、模型组、西药组、柴胡疏肝散精简方组(精简方组)。除空白组外,其余组大鼠每天腹腔注射利血平0.3mg/kg造模,空白组予等量生理盐水腹腔注射,每天上午注射1次,连续14d。西药组按照每天5mL(2mg)/kg灌胃给药,柴胡疏肝散精简方(柴胡9g,白芍15g,枳壳9g,香附9g)按每天5mL(8g)/kg灌胃给药,模型组及空白组予等量蒸馏水灌胃,腹腔注射利血平与灌胃给药同时进行,每日1次。对各组大鼠外观、形态、体质量及行为学等进行观察测量。对各组大鼠中缝核内儿茶酚胺进行检测。结果:与空白组比较,模型组大鼠体质量下降明显(P<0.05),旷场实验水平运动总距离及穿格数明显减少(P<0.01),强迫游泳实验不动时间明显增加(P<0.001);中缝核内去甲肾上腺素含量明显降低(P<0.01)。与模型组比较,西药组大鼠在给药后体质量增高,14d后具有统计学差异(P<0.05),大鼠水平运动总距离增加,差异有统计学意义(P<0.01),有增加大鼠穿格数趋势,但差异未见明显统计学意义(P>0.05),大鼠中缝核内去甲肾上腺素含量增加(P<0.05)。精简方组与模型组比较,大鼠体质量显著增加(P<0.05),旷场实验中水平运动总距离增加(P<0.001),强迫游泳实验中的不动时间减少(P<0.05),大鼠中缝核内去甲肾上腺素含量增加(P<0.05)。结论:柴胡疏肝散精简方具有一定的抗抑郁作用,可改善其抑郁样行为,并提高大脑中缝核内去甲肾上腺素的含量。 Objective:To observe the effects of Simplified Chaihu Shugan Powder(SCSP) on behavior of depressive-like rats and catecholamine in raphe nucleus caused by reserpine.Methods:Forty-eight SD rats were randomly divided into control group,model group,fluoxetine group,SCSP group,12 in each group.Reserpine was intraperitoneally injected for 14 days with 0.3 mg/kg to establish model in all the groups expect the control group.And the control group was injected with normal saline as same as the others.Meanwhile,the western medicine group and SCSP group were treated with 5 mL/kg of fluoxetine or SCSP.Both the control group and the model group were treated with water.All of them were used once a day.After 14 days,we recorded the appearance,weight,behaviors of TST and OFT.The monoamine catecholamines in raphe nucleus were detected by HPLC.Results:Compared with the control group,the weight decreased obviously(P<0.05),the total distance and grid numbers through in Open-Field were significantly reduced(P<0.01).The rest time in forced swimming test with statistical differences(P<0.001).The contents of 5-HT and NE decreased significantly in the raphe nucleus(P<0.01) in the model group.Compared with the model group,the downward of weight was not obvious,with statistical difference on day fourteenth(P<0.05) andthe level of exercise rats total distance increased.The difference was statistically significant(P<0.01) and rats cross number trend increased but the difference had no obvious statistical significance(P>0.05) in the western medicine group.Compared with the model group,the downward of weight was not obvious(P<0.05) and the total distance was apparently increased in Open-Field(P<0.001).The rest time in forced swimming test were decreased(P<0.05) in SCSP group.Conclusion:SCSP has certain antidepressant effect and can improve the depressive like behavior,but the brain monoamine catecholamines in the raphe nucleus has little effect.
