益精方对腺嘌呤法大鼠蛋白激酶A以及血清抑制素B表达的作用

Effect of Yijing Formula on Expression of Protein Kianse A and Serum Inhibin B of Adenine-Induced Infertile Rats

目的:观察益精方对腺嘌呤法诱导的肾阳虚型不育症大鼠睾丸蛋白激酶A(protein kianse A,PKA)以及抑制素B(inhibinB,INHB)表达的影响。方法:选用48只健康的SPF级Wistar雄性大鼠,2月龄,随机分为空白组(正常组)、阴性对照组(模型组)、阳性对照组(复方玄驹组)、实验组(益精方组)。每组大鼠12只。正常组以外,余下的各组大鼠均以1mL/(100g·d)剂量的腺嘌呤连续灌胃12d;从第13天开始,阴性对照组及空白组以相同剂量的生理盐水灌胃,实验组组用益精方汤剂灌胃,阳性对照组用复方玄驹胶囊水溶液灌胃,每天一次,连续20d。然后处死大鼠,摘取睾丸,采用RT—PCR技术测定PKAmRNA的表达;采用免疫组化技术测定大鼠睾丸组织中INHB的表达。结果:与空白组比较,阴性对照组PKAmRNA以及INHB的表达均显著降低(P<0.01)。与阴性对照组比较,实验组PKAmRNA以及INHB表达显著增加(P<0.05)。结论:益精方对腺嘌呤法诱导的肾阳虚大鼠睾丸组织中的PKAmRNA以及INHB表达具有明显的提高作用。

Objective:To observe the effect of Yijing Formula,a TCM compound,on the expressions of protein kianse A(PKA) and serum inhibin B(INHB) of infertile rats with kidney-yang deficiency pattern induced by adenine.Methods:Altogether 48 healthy male Wistar rats of SPF grade,two-month-old,were randomly divided into four groups:blank group,model group,Fufang Xuanju group and Yijing Formula group(n=12).Except rats in the blank group,rats in the other groups were intragastrically administrated with a dose of 1 mL/(100 g·d) adenine for 12 days.From the 13 th day on,rats in the blank and model groups were intragastrically treated with normal saline,and the rats in Fufang Xuanju group with Fufang Xuanju Capsule by aqueous gavage and the rats in Yijing Formula group with Yijing Formula,once a day for 20 consecutive days.After the experiment,the rats were sacrificed and their testis and epididymis were resected.The expressions of PKAmRNA were measured by RT-PCR,and the expressions of INHB were measured using immunohistochemical method.Results:Compared with the blank group,the expressions of PKAmRNA and INHB declined significantly in model group(P<0.01).Compared with the model group,the expressions of PKAmRNA and INHB increased in Yijing Formula group(P<0.05).Conclusion:Yijing Formula can obviously increase the expressions of PKAmRNA and INHB of kidney-yang deficiency pattern rats induce by adenine.
