新温肾生精饮对邻苯二甲酸二丁酯所致大鼠附睾组织氧化应激和精子损伤的影响

Effects of New Wenshen Shengjing Decoction on Epididymal Tissue Oxidative Stress and Sperm Injury in Rat Caused by Dibutyl Phthalate

摘要 目的:探讨新温肾生精饮对邻苯二甲酸二丁酯(dibutyl phthalate,DBP)所致大鼠附睾组织氧化应激和精子损伤的影响,为临床用药提供可行依据。方法:将青春期SD级雄性大鼠90只分为空白组15只和造模组75只。造模组大鼠给予DBP灌胃造模,60只大鼠成功造模,将其分为模型组、新温肾生精饮高剂量组[12 g·(kg·d)-1]、新温肾生精饮中剂量组[8 g·(kg·d)-1]、新温肾生精饮低剂量组[4 g·(kg·d)-1],每组各15只。空白组与模型组给予等量生理盐水灌胃。采用精子质量分析系统检测精子浓度、活力及畸形率,使用比色法检测丙二醛(malondialdehyde,MDA)、谷胱甘肽(glutathione,GSH)和谷胱甘肽过氧化物酶(glutathion peroxidase,GSH-px)水平,测量生精小管直径(diameter of seminiferous tubules,STD)和生精上皮高度(seminiferous epithelium height,SEH),检测卵泡刺激素(follicle-stimulating hormone,FSH)、大鼠促黄体生成素(luteinizing hormone,LH)和睾酮(testosterone,T)。结果:空白组与模型组的精子浓度、活力及畸形率、MDA、GSH、GSH-px、生精小管直径、SEH、FSH、LH、T水平比较,差异均有统计学意义(P <0. 05);模型组与新温肾生精饮各剂量组的精子浓度、活力及畸形率、MDA、GSH、GSH-px、STD、SEH、FSH、LH、T水平比较,差异均有统计学意义(P <0. 05)。新温肾生精饮高剂量组精子浓度、活力及畸形率、MDA、GSH、GSH-px、STD、SEH、FSH、LH、T水平等指标均优于新温肾生精饮低剂量组和中剂量组,差异有统计学意义(P <0. 05)。结论:DBP会造成睾丸损伤,导致精子浓度、活力降低,畸形率上升,MDA、FSH升高,GSH、GSH-px、STD、SEH、LH、T水平降低;新温肾生精饮能够改善睾丸结构,修复附睾组织氧化应激损伤和精子损伤,提高生殖能力。 Objective: To investigate the effects of New Wenshen Shengjing Decoction on epididymal tissue oxidative stress and sperm injury in rat caused by dibutyl phthalate and provide a feasible basis for clinical drug use. Methods: 90 SD male rats were divided into blank group and model group,with 15 in blank group and 75 in model group. Rats in model rats were given DBP for intragastric administration. After 60 rats were successfully modeled,they were divided into model group,New Wenshen Shengjing De- coction high-dose group of 12 g( kg·d)^-1,New Wenshen Shengjing Decoction medium-dose group of 8 g( kg·d)^-1, and New Wenshen Shengjing Decoction low-dose group of 4 g( kg·d)^-1, 15 in each group. The blank group and the model group were given the same amount of normal saline. Sperm quality analysis system was used to detect sperm concentration,vigor and deformity rate. The colorimetric method was used to detect the levels of malondialdehyde ( MDA) , glutathione ( GSH) and glutathion peroxidase ( GSH-px) , and the diameter of seminiferous tubules ( STD) , seminiferous epithelium height ( SEH) , follicle-stimulating hormone ( FSH) , rat luteinizing hormone ( LH) and testosterone ( T) were measured. Results: There were significant differences in sperm concentration,vigor and deformity rate,MDA,GSH,GSH-px,diameter of seminiferous tubules,SEH,FSH,LH and T levels between the blank group and the model group ( P < 0. 05) . There were significant differences in sperm concentration,vigor and deformity rate,MDA,GSH,GSH-px,STD,SEH,FSH,LH and T levels between the model group and New Wenshen Shengjing Decoction group ( P < 0. 05) . The sperm concentration,vigor and deformity rate,MDA,GSH,GSH-px,STD,SEH,FSH,LH,T levels in the high-dose group of New Wenshen Shengjing Decoction were better than those in the low-dose group and the middle-dose group. The differences were statistically significant ( P < 0. 05) . Conclusion: DBP can cause testicular damage,resulting in decreased sperm concentration,vitality,increased sperm deformity,increased MDA,FSH,and decreased levels of GSH,GSH-px,STD,SEH, LH,and T. New Wenshen Shengjing Decoction can improve testicular structure,repair oxidative stress damage and sperm damage in epididymis,and improve reproductive ability.
