摘要
目的探索坐骨神经分支选择性损伤(sparednerveinjury,SNI)模型中小胶质细胞抑制剂氟代柠檬酸(Fluorocitrate,FC)对大鼠痛敏的影响。方法取成年雄性SD大鼠24只,随机均分为四组:假手术组(Sham组)、模型组(SNI组)、SNI+生理盐水组(SNI+NS组)和SNI+氟代柠檬酸(SNI+FC组)。用VonFrey纤维丝测定四组大鼠50%缩足阈值(50%mechanicalwithdrawalthreshold,50%MWT)的变化;SNI+FC组和SNI+NS组于术后8天开始分别给予FC或NS,隔天1次,共3次,术后13d取材,利用Western-blot技术检测各组脊髓GR蛋白的表达情况。结果与Sham组相比,SNI组大鼠术后50%MWT显著降低(P<0.05);与SNI+NS组相比,SNI+FC组大鼠术后50%MWT显著增加(P<0.05);Western-blot显示,术后SNI+FC组大鼠GR的表达增加,与SNI+NS组相比差异均有统计学意义(P<0.05)。结论SNI大鼠模型可成功模拟神经病理性疼痛;与SNI+NS组相比,Western-blot显示,SNI+FC组大鼠术后GR的蛋白水平显著增加(P<0.05),表明鞘内注射氟代柠檬酸可缓解坐骨神经分支选择性损伤大鼠神经病理性痛。
Objective To evaluate the effects of Fluorocitrate( FC),a microglial inhibitor,on the mechanical pain sensitivity induced by the spared nerve injury( SNI) model rats. Methods Twenty-four male Sprague-Dawley adult rats were randomly divided into four groups: Sham,SNI,SNI + NS and SNI + FC. The 50% mechanical withdrawal threshold( 50% MWT) were measured by Von Frey filament. FC or NS were injected intrathecally from the 8 th day to the13 th day,every other day,a total of three times. Detected the expression of GR protein in SNI + FC and SNI + NS group on 13 th day by Western-blot. Results Compared with the Sham group,50% MWT of SNI group was significantly decreased( P < 0. 05). Compared with SNI + NS group,50% MWT of SNI + FC group increased significantly( P < 0. 05). Compared with the SNI + NS group,Western-blot in SNI + FC group showed the expression of GR was increased( P < 0. 05). Conclusion The SNI model could successfully replace the neuropathic pain. Compared with the SNI + NS group,Western-blot showed that the level of GR protein in SNI + FC group was significantly increased( P < 0. 05).
