摘要 设计并制作了一种可以对人体心肌钙蛋白Ⅰ浓度进行电化学检测的集成微带阵列电极微流控芯片,应用于急性心肌梗塞的早期诊断。芯片由一片聚二甲基硅氧烷(PDMS)沟道片和一片玻璃电极片组成,在PDMS沟道片上制作了可以固定心肌钙蛋白Ⅰ的微通道,在玻璃片上集成了用于心肌钙蛋白Ⅰ浓度检测的微带阵列电极。包被抗体通过蛋白G修饰层固定于硅烷化的PDMS沟道内表面,利用双抗体夹心法得以实现浓度检测。通过循环伏安法获得的氧化电流峰值正比于酶反应产物,即对氨基磷酸苯酚(PAP)的浓度。心肌钙蛋白I的理论检出限为0. 4 ng/m L,线性检测范围为1~1 000 ng/m L,检测时长为8 min。 A micro-fluidic chip with integrated microband array electrode is designed and fabricated for the detection of human cardiac troponin Ⅰ(cTnI)in the early diagnosis of acute myocardial infarction.The chip consists of a polydimethylsiloxane(PDMS)channel plate and a glass electrode plate.One micro-channel to immobilize cTnI coating antibody is integrated on the PDMS channel plate.Microband array electrode for the concentration detection of cTnI is integrated on the glass electrode plate,cTnI coating antibody is immobilized onto the internal surface of the PDMS channel on which protein G layer is generated by silanization.The detection experiments are performed with the double antibody sandwich method.Cyclic voltammograms are obtained by the oxidation peak current proportionally to the concentration of enzymatic product,p-aminophenol (PAP).The theoretical limit of detection is 0.4ng/mL within 8min,the linear detection range is 1~1000ng/mL.
