摘要
目的探讨微小RNA-101及转化生长因子-β信号通路在肝纤维化中的作用。方法选取2013年1月至2018年1月间宜宾市第一人民医院收治的40例原发性肝癌合并肝纤维化患者手术切除的肝纤维化组织为研究组,另选取40例因肝转移癌患者行肝脏切除的肝正常组织为对照组。采用免疫组化法分别检测肝组织中转化生长因子-β(TGF-β1)和平滑肌α-肌动蛋白(α-SMA)表达水平,采用qRT-PCR检测肝组织中miR-101表达情况,分析TGF-β1和α-SMA与miR-101的相关性。结果肝纤维化组织中TGF-β1蛋白表达阳性率高于正常肝组织,差异有统计学意义(P <0. 05);两组标本中α-SMA表达阳性率比较,差异无统计学意义(P> 0. 05)。TGF-β1和α-SMA蛋白在肝纤维化组织中的阳性表达随肝纤维化程度的加重而增强,呈正相关性,差异均有统计学意义(均P <0. 05)。TGF-β1和α-SMA蛋白阳性的表达率呈正相关,差异有统计学意义(P <0. 01)。肝纤维组织miRNA-101水平低于正常肝组织,差异有统计学意义(P <0. 05)。miR-101与TGF-β1和α-SMA蛋白表达呈负相关,差异均有统计学意义(均P <0. 01)。结论 miRNA-101下调在肝纤维化过程中起重要作用,可能通过调控TGF-β1而参与肝纤维化。
Objective To explore the roles of miR-101 and transforming growth factor-β( TGF-β1) signalling pathway in liver firosis. Methods Forty patients with primary liver complicated with fibrosis who received treatment and underwent surgery at The First People's Hospital of Yibin from January 2013 to January 2018 were selected the observation group. Another 40 with metastatic liver cancer who had normal liver tissues resected were selected as the control group. The expression of TGF-β1、α-smooth muscle actin( α-SMA) was detected by immunohistochemistry. The expression level of miR-101 in fibrotic liver tissues was detected by quantitative real time polymerase chain reaction( qRT-PCR). The correlation between TGF-β1 and α-SMA and miR-101 in the liver fibrosis was analyzed.( P<0. 05). Results TGF-β1 expression was much higher in liver fibrosis group than in group with normal liver tissues( P<0. 05). Therer was no significant difference in the positive expression of α-SMA between the two groups( P<0. 05). The increased expression of TGF-β1 and α-SMA were closely correlation to the degree of hepatic fibrosis( all P<0. 05). Expression of TGF-β1 was significantly correlated with the expressionα-SMA( P<0. 05). miR-101 was less expressed in liver firosis than in normal liver tissues( P<0. 05). The expression of miR-101 was negatively correlated with the expression of TGF-β1 and α-SMA( all P<0. 01). Conclusion Downregulation of miR-101 may play an important role in liver firosis by controlling TGF-β1.
