原花青素对喉癌TU686细胞增殖、凋亡的影响 被引量:4

Effects of procyanidins on the proliferation and apoptosis of laryngeal carcinoma TU686 cells

摘要 目的探讨原花青素对喉癌TU686细胞增殖、凋亡的影响及可能机制。方法采用20、40、60、80μg/ml的原花青素作用于喉癌TU686细胞,MTT方法检测细胞增殖情况,流式细胞术检测细胞凋亡,计算半数抑制浓度。用半数抑制浓度的原花青素作用于喉癌TU686细胞,Western blot方法与real time PCR方法分别检测磷酸化的蛋白激酶B(p-Akt)与磷酸化的信号转导与转录激活因子3(p-STAT3)的蛋白与m RNA表达水平。结果原花青素能够呈时间、浓度依赖地抑制喉癌TU686细胞的增殖,半数抑制浓度为60μg/ml。采用半数抑制浓度的原花青素处理后,喉癌TU686细胞凋亡率呈时间依赖性地升高,细胞p-Akt与p-STAT3的蛋白与m RNA表达水平均显著降低,P<0.01。结论原花青素能够抑制喉癌TU686细胞的增殖并促进凋亡,下调p-Akt与p-STAT3的表达可能是其作用机制之一。 Objective To investigate the effects of procyanidins on the proliferation and apoptosis of laryngeal carcinoma TU686 cells and its possible mechanism. Methods The proanthocyanidins of 20, 40, 60 and 80 g/ml were used to treat TU686 cells of laryngeal carcinoma. The proliferation of the cells was detected by MTT method. The apoptosis was detected by flow cytometry, and the inhibitory concentration was calculated. The expression of phosphorylated protein kinase B(p-Akt) and phosphorylated signal transduction(p-STAT3) protein and mRNA was detected by Western blot and real time PCR method respectively after TU686 cells was treated by median inhibitory concentration of Proanthocyanidin. Results Proanthocyanidin could inhibit the proliferation of laryngeal carcinoma TU686 cells in a time and concentration dependent manner, its median inhibitory concentration was 60μg/ml. The apoptosis rate of TU686 cells in larynx cancer cells was increased in a time dependent manner with a median inhibitory concentration of proanthocyanidins, and the protein and mRNA expression levels of p-Akt and p-STAT3 decreased significantly after TU686 cells was treated by median inhibitory concentration of Proanthocyanidin. Conclusion Procyanidins might be a target for treatment of laryngeal carcinoma in clinic.
