摘要
目的:探讨Klotho基因rs1207568与新疆地区维吾尔族特发性草酸钙肾结石的相关性。方法:选择400例维吾尔族草酸钙肾结石患者(病例组)和410例维吾尔族健康体检者(对照组),应用SNaPshot方法对Klotho基因rs1207568进行基因检测,并分析其与特发性草酸钙肾结石发病的相关性以及对血脂、血生化的影响。结果:对照组的基因型分布均符合Hardy-Weinberg平衡。病例组和对照组rs1207568基因型比较差异有统计学意义(P<0.01),且病例组携带等位基因C的风险高于对照组(OR=1.295,95%CI:1.019~1.645,P<0.05)。CC基因型患者的血钙、血磷、血氯、尿素氮、尿酸均高于CT+TT基因型,差异有统计学意义(P<0.05),而二者血镁、血钾、血钠水平比较差异无统计学意义。在血脂方面,二者甘油三酯(TG)、总胆固醇(TC)比较差异有统计学意义(P<0.05)。结论:Klotho基因rs1207568在维吾尔族人群中与特发性草酸钙肾结石发病风险显示有相关性,rs1207568C等位基因是其危险因素。
Objective:To investigate the correlation between Klotho gene and rs1207568 and Uygur special calcium oxalate kidney stone in Xinjiang area.Method:The 400 Uygur patients with calcium oxalate nephrolithiasis and 410 Uygur health examiners were examined by SNaPshot method to detect the Klotho gene rs1207568,and its correlation with calcium oxalate kidney stones and its effects on blood lipids and blood biochemistry were analyzed.Result:The genotype distribution of the Uygur control group was in line with the Hardy-Weinberg balance.The genotype of rs1207568 loci in Uygur case group and control group had significant statistical difference(P<0.01).Moreover,the risk of carrying allele C in Uygur case group was higher than that in Uygur control group(OR=1.295,95%CI:1.019-1.645,P<0.05).The CC genotype of the Klotho gene rs1207568 loci was compared with the CT+TT genotype.The patients were significantly higher than the latter in blood calcium,blood phosphorus,blood chlorine,urea nitrogen,uric acid(P <0.05).There was no significant difference in blood magnesium,blood potassium or blood sodium levels.TG and TC had statistical significance(P <0.05).Conclusion:The Klotho gene rs1207568 is associated with the risk of idiopathic calcium oxalate kidney stones in the Uygur population,and the C allele of rs1207568 loci is a risk factor.
