茄科植物A622基因的起源与进化

The Origin and Evolution of A622 Gene in Solanaceae

摘要 A622基因在烟草砒啶类生物碱生物合成中起重要作用,其在茄科植物中的起源和进化还未有报道。本研究鉴定得到该基因含有NmrA-like家族结构域,且该结构域几乎覆盖A622基因全长。通过搜索栽培烟草、番茄、辣椒,以及咖啡和拟南芥基因组,得到了92条NmrA-like家族成员。将A622基因与拟南芥基因单独聚类,发现拟南芥基因大体分为两大枝;与A622聚在一起的枝中,含有已鉴定功能的PR基因。通过CLANS聚类,得到包含已知A622基因和拟南芥PR基因的46条基因。采用最大似然法,将此类基因进行进一步聚类,得到一个包含A622基因的亚家族,该亚家族含有1条拟南芥基因、1条咖啡基因、2条烟草基因以及3条番茄基因。本研究为将来进一步研究A622在茄科植物中的功能提供基础;结合对拟南芥基因(AT4G39230)的功能及关键位点鉴定,将能进一步阐明茄科植物A622基因的起源与进化。 A622 gene plays an important role in pyridine alkaloid biosynthesis in tobacco, whose origin and evolution in Solanaceae has not been reported yet. In this study, we identified that the A622 gene contained NmrA-like family domain, which almost spans the full-length of A622 peptides. Through searching the genome of cultivated tobacco, tomato, chili, coffee and Arabidopsis, 92 NmrA-like family members were obtained. By clustering A622 gene and Arabidopsis thaliana gene separately, it was found that Arabidopsis gene was divided into two branches, and the branch with A622 contained the identified PR gene. Through CLANS clustering, 46 genes were obtained which include known A622 gene and Arabidopsis PR gene. By using maximum likelihood method, further phylogenetic analyses identified a subfamily containing tobacco A622 gene, which included 1 Arabidopsis gene, 1 coffee gene, 2 tobacco genes and 3 tomato genes. This study might provide foundations for the future studies of A622 in Solanaceae. Combined with functional analysis and key loci identification of the Arabidopsis gene (AT4G39230), the origin and evolution of A622 gene might be further characterized.
