大麻素2型受体调控自噬在脓毒症治疗中的研究进展 被引量:2

Research progress of cannabinoid receptor 2 in the treatment of sepsis by regulating autophagy

摘要 脓毒症是因感染引起的宿主反应失调而导致危及生命的器官功能障碍;由脓毒症引起的循环和细胞/代谢异常的高死亡率状态称脓毒症休克。全球每年有数百万人罹患脓毒症,其中1/4或更多的患者死亡。大麻素2型受体(CB2R)属于G蛋白偶联受体,主要表达在免疫组织中。研究表明,CB2R参与了多种疾病的炎症反应过程。细胞自噬是真核生物中进化保守的对细胞内物质进行周转的重要过程。越来越多的研究表明自噬参与了细胞的免疫过程,通过清除病原体和负向调节Nod样受体蛋白-3(NLRP3)炎症小体调控免疫过程。大麻素通过调控自噬发挥抗炎和免疫调节作用,使它成为治疗脓毒症的新靶点,为脓毒症的治疗带来了新希望。 Sepsis is life-threatening organ dysfunction caused by a dysregulated host response to infection.Septic shock is a subset of sepsis with circulatory and cellular/metabolic dysfunction associated with a higher risk of mortality. Sepsis and septic shock are major healthcare problems, affecting millions of people around the world each year, and killing as many as one in four(and often more). Cannabinoid receptor 2(CB2 R) belongs to G protein coupling receptor, which is mainly expressed in immune tissue.Studies have shown that CB2 R is involved in inflammatory responses to a variety of diseases. Autophagy is an important process in eukaryotic organisms that is evolutionarily conserved for turnover of intracellular materials. Several lines of evidence strongly suggest that autophagy is involved in the im-mune process, which is regulated by removing pathogens and negatively regulating the Nod-like receptor pyrin domain 3(NLRP3) inflammasome. Cannabinoids exert anti-inflammatory and immunomodulatory effects by regulating autophagy, making it a new target for the treatment of sepsis and bringing new hope for sepsis.
