摘要
目的采用回流提取的方法,利用高效液相色谱-质谱联用技术(HPLC-MS/MS)测定瓜蒌-薤白中槲皮素的含量。方法用AgilentC18(4.6mm×250mm,5 m)色谱柱,流动相为甲醇-0.01%甲酸的水溶液,检测波长230nm,柱温30℃,进样量3 L。采用电喷雾离子源,正离子模式,多反应监测(MRM)扫描方式。结果槲皮素的质量浓度在52.5~1575ng/mL范围内(r=0.99375)与峰面积分值呈良好的线性关系,平均加样回收率为96.59%(RSD=0.99%)。结论该方法重复性好、灵敏度高、专一性强,可以用作瓜蒌-薤白化学成分的含量测定。
Objective To detemine the content of quercetin in Trichosanthes kirilowii and Allium macrostemon by high performance liquid chromatography-mass spectrometry (HPLC-ms/ms)with reflux extraction method.Methods Agilent 18 (4.6mm×250mm,5μm)column was used.The mobile phase was methanol-0.01%formic acid aqueous solution.The detection wavelength was 230nm,the column temperature was 30℃,and the injection volume was 3μL.Results The concentration of quercetin in the range of 52.5-1575ng/mL(r=0.99375)showed a good linear relationship with the peak area score.The average recovery rate of quercetin was 96.59%(RSD=0.99%).Conclusion The results showed that this method has good reproducibility, high sensitivity,high specificity,can be used for content determination in Trichosanthes kirilowii and Allium macrostemon.
