自噬相关蛋白mTOR、LC3-Ⅱ及HIF-1α与PM/DM的相关性

The relationship of autophagy related protiens m TOR,LC3-Ⅱ and hypoxia induced factor-1α with the polymyositis and dermatomyositis

目的检测自噬相关蛋白m TOR和LC3-Ⅱ及HIF-1α在PM/DM患者肌肉组织中的表达水平,进一步探讨自噬相关蛋白m TOR、LC3-Ⅱ及HIF-1α在PM/DM发病机制中的作用及意义。方法应用免疫组化法测定PM/DM患者肌肉组织中m TOR、LC3-Ⅱ、HIF-1α的表达水平,并行相关研究。结果 PM和DM组肌肉组织中m TOR蛋白的阳性表达水平均低于Control组(P<0.05); PM和DM组肌肉组织中LC3-Ⅱ蛋白的阳性表达水平均高于Control组(P<0.05); PM组和DM组中m TOR与LC3-Ⅱ蛋白的表达水平均呈负相关(P<0.05); DM组中HIF-1α蛋白的阳性表达水平高于PM组和Control组(P<0.05); PM和DM组MDAAT+VAS评分与LC3-Ⅱ蛋白表达水平均呈正相关(P<0.05)。差异均有统计学意义。结论自噬相关蛋白m TOR、LC3-Ⅱ可能参与了PM/DM的发病,LC3-Ⅱ与疾病的活动度有关; HIF-1α参与了DM的发病。

Objective To determine the relationship of autophagy related proteins such as mammalian target of rapamycin( m TOR),microtubule-associated protein light chain( LC3-Ⅱ) and the level of hypoxia induced factor-1α( HIF-1α) with the pathogenesis of polymyositis( PM) and dermatomyositis( DM). Method The expressions of m TOR,LC3-Ⅱ and HIF-1α in the muscle tissues of patients with PM or DM were detected by immunohistochemical method.Results Less m TOR was expressed in PM and DM than that in control( P<0.05); the expression of LC3-Ⅱ was higher in DM and PM than that in control( P<0.05) and HIF-1α was expressed higher in DM than those in PM and control( P<0.05); m TOR and LC3-Ⅱ were negatively related and LC3-Ⅱ was positively related with MDAAT+VAS score.Conclussion Autophagy related protiens m TOR and LC3-Ⅱ may be involved in the pathogenic process of PM/DM; LC3-Ⅱ may related with the activity of the PM and DM and HIF-1α may take part in the pathogenic process of DM.
