摘要
目的:建立四物汤UPLC指纹图谱的分析方法,并应用于传统煎液与中药配方颗粒调配剂的对比。方法:采用Luna Omega C18(150×2.1mm,1.6μm)色谱柱,流动相为乙腈-0.1%磷酸溶液(梯度洗脱),流速为0.2mL/min;使用230nm和277nm两个波长进行采集;运用"中药色谱指纹图谱相似度评价系统"软件对样品进行相似度评价。结果:在230nm条件下,四物汤中检测出10个白芍的特征共有峰;在277nm条件下,检测出当归、川芎和熟地黄23个共有峰;中药配方颗粒调配剂指纹图谱与传统煎液对照谱图对比,相似度良好,在0.957~0.991之间。结论:所建立的四物汤UPLC双波长指纹图谱,其方法精密度、重复性和稳定性良好;通过对多个四物汤传统煎液样本与中药配方颗粒调配剂的对比分析,结果显示两者的物质成分无显著差异。
Objective: To establish the method for UPLC fingerprint of Siwu Decoction, and apply it in the contrast Traditional Decoction with themix of formula granules of Siwu Decoction. : Method: The separation was developed on a Luna Omega C 18 column(150×2.1mm, 1.6μm) by gradientelution with ACN-water (containing 0.1% phosphoric acid ) as the mobile phase, flow rate at 0.2mL/min, detection wavelength at 230nm and 277nm.And The Traditional Decoction was evaluated by similarity assay with the software of "Chinese medicine chromatographic fingerprint similarity eval-uation system" (2012.130723). : Result: 10 common peaks originated from Paeoniae radix Alba were separated from Siwu Decoction at 230nm; 23common peaks were separated at 277nm, which stemed from Angelicae sinensis radix, Chuanxiong radix and Rehmanniae radix praeparata; the sim-ilarity between the mix of formula granules of Siwu Decoction fingerprint and reference fingerprint of Traditional Decoction was good in 0.957~0.991. : Conclusion: The dual-wavelength UPLC fingerprint method of Siwu Decoction was precise, re-producible and stable; the results show thatthere is no significant difference between traditional decoction and mix of formula granules of Siwu Decoction.
