圈养非人灵长类乳酸杆菌的分离筛选和功能鉴定 被引量:2

Isolation,screening and functional identification of Lactobacillus from captive non-human primates

摘要 乳酸杆菌对于动物的肠道健康有着非常重要的作用。为研究圈养非人灵长类动物肠道乳酸杆菌,无菌采集不同品种的表观健康的圈养灵长类动物新鲜粪便,进行乳酸杆菌的分离培养及纯化,并通过耐酸耐胆盐试验、细胞黏附试验和对大肠杆菌抑菌试验,对其进行生物学特性研究。结果显示:共分离得到17株乳酸杆菌,有3株菌Lac5-2、Lac5-4、Lac6-2在pH 3.0和胆盐浓度0.5%时表现出良好耐酸耐胆盐性能,且无耐药性。其中菌株Lac6-2黏附性及对大肠杆菌的黏附抑制性均较好,黏附率为16.2%(MOI=100),黏附抑制率为85.5%(MOI=10)。经16S rDNA测序,Lac5-2、Lac5-4、Lac6-2均为罗伊氏乳酸杆菌。最终确定菌株Lac6-2为候选优势益生菌株。该研究为利用乳酸杆菌防治圈养野生动物消化道疾病提供基础。 Lactic acid bacteria plays an important role in the intestinal health of animals.To study the intestinal lactic acid bacteria in Captive Non-human Primates,the stools from healthy Captive Non-human Primates were collected sterilely,then the isolation and purification of lactic acid bacteria were performed,and their biological characteristics were identified by the acid and bile tolerance test,adhesion test and antimicrobial test.The results showed that 17 strains of Lactobacillus were isolated from the feces of captive non-primate,three isolates(Lac5-2,Lac5-4 and Lac6-2) demonstrated good performance at pH 3.0 and bile 0.5%(w/v),respectively.they were not drug-resistant.One isolate(Lac6-2) from squirrel monkeys was found to have good adhesion and excellent antimicrobial ability.The adhesion rate was16.2%(MOI=100),while the inhibition rate of adhesion was 85.5%(MOI=10).These three isolates were identified as Lactobacillus reuteri by 16 S r DNA sequencing.Isolate Lac6-2 was considered as the best probiotic candidate.This study provided necessary and important theoretical guidance for further the treatment of digestive tract diseases in captive wild animals by Lactobacillus.
